Laboratory exercise #5 ANTI-STREPTOLYSIN O TITER I. Results /Observation Table 1.

Result for ASOT Type of Test Qualitative Quantitative

Result Negative: no agglutination is observed. Negative: no agglutination is observed.

II.

Discussion The ASO test is primarily ordered by itself or along with an anti-DNase B to help determine whether a person has had a recent streptococcal infection. In most cases, strep infections are identified and treated with antibiotics and the infections resolve. In cases where they do not cause identifiable symptoms and/or go untreated, however, post-streptococcal complications (sequelae), namely rheumatic fever and glomerulonephritis, can develop in some patients, especially young children. The test, therefore, is ordered if a person presents with symptoms suggesting rheumatic fever or glomerulonephritis and has had a recent history of sore throat or a confirmed streptococcal infection

Guide Questions: 1. What factors may cause false positive aSOT result? Describe each. Table 2. Factors of false positive result in aSO assay Tuberculosis Microbial contamination of the serum with Bacillus and Pseudomonas, presence of these could trigger aSO antibodies react and act upon it. Lipemia This may cause non specific agglutination. Both elevated Beta-lipoprotein and cholesterol level can neutralize the ASOT. Lapsed Time Delayed readings (after 2 minutes) may cause agglutination leading to false positive result. Liver Disease Malfunction causes malabsorption of fats makes the blood specimen from which serum is obtained chylous. 2. Why time is very important in the interpretation of aSOT test? 3. How important is the sensitivity and specificity of an aSo assay for the management of rheumatic heart disease and other conditions associated with streptococcus infection?
