Lab Name: Staining Lab-Simple Stain Purpose: The difference in shape of microorganisms is a very important tool in understanding the specific characteristics of a microbe. The simple staining method helps us to see the different morphologies of microorganisms. They generally fall into three categories: Bacillus, Cocci and Spirilla. The purpose of this lab is to illustrate how the simple stain helps to illuminate the morphology of microorganisms as well as show how to actually perform the simple staining technique effectively. Materials: -Tryptic soy agar Petri dishes -Clothespins -1% Methylene Blue Organisms: -Unknown 1 Procedure: 1. 2. 3. 4. 5. Label slides Unknown 1 and Unknown 2. Cover smear with 1% Methylene Blue Allow stain to react with smear for 30-45 seconds then allow excess to drip off Rinse slide by dipping briefly in a beaker of water Examine stained cells under microscope
