ABSTRACT
The existence of NH4NO3 and CaCl2 in MS medium used in carnation tissue culture has high effect on response
of explant growth cultured in vitro. The response of explant can be a normal and healthy growth or hyper-
hydritised-growth. The study aimed to know concentration changes of two macro elemen on hyperhydri-
tised-explant growth of carnation. The experiment was conducted at Tissue Culture Laboratory, IPPT, Faculty of Agriculture,
Universiti Putra Malaysia from January to August 2002. Factorial experiment was arranged using Randomized Complete Block
Design with four replications. First factor was concentration of NH4NO3 of (1) normal concentration, (2) ½ of normal concen-
tration, (3) ¼ of normal concentration and (4) without NH NO . Second factor was concentration of CaCl of (1) 1.5 of normal

concentration, (2) normal concentration, (3) ½ of normal concentration and (4) without CaCl2. Results of the study indicated that
concentration changes of NH4NO3 dan CaCl2 gave high effect on hyperhydritised-explant growth. Half strength of NH4NO3 and
CaCl2 from thier normal concentration was the appropriate concentration in reducing hyperhydricity incidence down to 67%
and increasing the highest leaf chlorophyll content up to 34% ( from 0.088 mg/mg to 0.134 mg/mg). Normal concentration of
CaCl2 without NH4NO3 presence in MS medium was the most suitable combination in lowering water content and enhancing
dry weigth of shoots. The existence of half-concentration of NH4NO3 without CaCl2 in the MS medium induced high axillary
shoot proliferation (8.6 shoot per explant). While the highest averages of shoot height were obtained on MS medium with normal
concentration of NH4NO3 without CaCl2.
