Lab Practical: Lab 2 Review

The Microscope

Objectives
o Scanning Power Objective (X40 Magnification): The first objective to be used for sample
observation.
o Low Power Objective (X100 Magnification)
o High Power Objective (X400 Magnification)
Revolving Nosepiece: by rotating the nosepiece, the user can change the objective in use.
Eyepiece (ocular): can be adjusted to match the pupil distance of the user for stereo vision. The
eyepiece has a magnification power of X10.
Condenser: concentrates the light to the specimen and the iris diaphragm.
Iris diaphragm: moving the lever of the diaphragm regulates the amount of light coming from
the light source.
Course focusing knob: should only be used when the scanning power objective is in place.
Magnification and Resolution
o The resolution is the ability of the microscope to distinguish two points as separate.

With increased magnification and resolution, the viewer will have a smaller surface area
of the specimen to analyze.
