Abstract
Wepreviouslyreportedsixneuroprotectivedecursinolderivatives,coumarinsfromAngelica
gigas(Umbelliferae)roots.Toelucidatetheactionpatternsofdecursinolderivatives,we
investigatedtheneuroprotectiveeffectsofdecursinolanddecursin,whichshowedhighly
significantactivityandweremajorconstituentsofA.gigas,usingprimaryculturesofratcortical
cellsinvitro.Atconcentrationsof0.110.0microM,bothdecursinolanddecursinexerteda
significantneuroprotectiveactivitypretreatmentandthroughouttreatment.Inaddition,decursin
hadaneuroprotectiveimpactintheposttreatmentparadigmimplyingthatdecursinmight
possessdifferentactionmechanismsfromthatofdecursinolintheprotectionofneuronsagainst
glutamateinjury.Bothdecursinolanddecursineffectivelyreducedtheglutamateinduced
increasedintracellularcalcium([Ca(2+)](i))incorticalcells,suggestingthatthesetwocoumarins
mayexertneuroprotectionbyreducingcalciuminfluxbyoveractivationofglutamatereceptors.
Thissuggestionwassupportedbytheresultthatdecursinolanddecursinprotectedneurons
againstkainicacid(KA)inducedneurotoxicitybetterthanagainstthatinducedbyNmethylD
aspartate(NMDA).Moreover,bothdecursinolanddecursinsignificantlypreventedglutamate
induceddecreasesinglutathione,acellularantioxidant,andglutathioneperoxidaseactivity.In
addition,bothcompoundsefficientlyreducedtheoverproductionofcellularperoxidein
glutamateinjuredcorticalcells.Theseresultssuggestedthatbothdecursinolanddecursin
protectedprimaryculturedratcorticalcellsagainstglutamateinducedoxidativestressbyboth
reducingcalciuminfluxandactingonthecellularantioxidativedefencesystem.Moreover,
decursinisconsideredtoprobablyhaveadifferentactionmechanismfromthatofdecursinolin
protectingcorticalcellsagainstglutamateinjury.
