TEST PROCEDURE:
The LINEAR ARRAY HCV Genotyping Test is an in vitro test to be used for determining the genotype (GT1-GT6) of
Hepatitis C Virus. The test utilizes reverse transcription of target RNA to generate complementary DNA (cDNA),
amplification of target (cDNA) by the Polymerase Chain Reaction (PCR), and nucleic acid hybridization for the
genotyping of HCV RNA in human serum or EDTA plasma.
The LINEAR ARRAY HCV Genotyping Test is based on five major processes:
Specimen preparation; reverse transcription of the target RNA to generate complementary DNA (cDNA); PCR
amplification of target cDNA using HCV specific complementary primers; hybridization of the amplified products to
oligonucleotide probes specific to the target (s); and detection of the probe-bound amplified products by the
colorimetric detection.
Comments:
Genotyping of HCV is dependent on the number of virus particles present in the specimen and may be affected by
specimen collection methods, patients factors (i.e. age, presence of symptoms), and/or stage of infection.
NOTE:
HCV Genotyping tests carried out in this laboratory can not be compared with the tests performed in the other
laboratories, due to differences in specimen storage, transportation, specification of assay used and quality control
standards.
