1) Prepare Stock Pararosaniline Solution 0.1 gm of Pararosaniline in distilled water. Stand for 48 hours then filter and content in 50 ml volumetric flask and make up with distilled water. 2) Prepare Solution 1 M Acetate acetic acid buffer stock solution with pH of 4.79 by dissolving 13.61 gm of sodium Acetate triydrate in distilled water in 10 ml volumetric flask. Add 5.7 ml of glacial Acetic Acid and dilute to volume with distilled water. 3) Prepare 1 ml of the stock PRA solution to 100 ml volumetric flask and dilute to volume with distilled water. 4) Transferred 5 ml aliquot of the diluted PRA solution from 2 into a 50 ml volumetric flask. Add 5 ml of 1 m acetate acetic acid buffer solution from 1 and dilute the mixture to volume with distilled water and let the mixture stand for 1 hour. 5) Take the absorbance of above solution at 540 nm. nm 500 510 520 530 540 550 560

nm when assayed in a buffered solution of 0.1 M sodium acetate acetic acid against a distilled water reference. % PRA=Absorbance x 21.3/0.1 where, 21.3 = spectrophotometer having a spectral band with of less than 15 nm and a path length of 1cm. 0.1 = weight of PRA for preparing stock solution
