The Posides and Tecans are the janitorial staff of the factory.

The Tecans were the

first generation staff (topoisomerase I) and the Posides are the second generation
(topoisomerase II). Eto and Teni are rednecks. Eto is always scratching his testicles
(testicular cancer) and Teni is always talking about having kids (Pediatric ALL).
Topo and Irin Tecan are the old couple. Between them, Topo has had Colorectal and
Small-cell lung cancer and Irin has had ovarian cancer. Mr. DR, the 1st (MDR-1 is
their boss just like for the Vinca twins and the Taxanes).
The DNA Intercalating team is basically the quality checkers, they intervene by
intercalating themselves into DNA and causing DNA breaks before the DNA product
gets to the RNA processing phase. They are the attractive group in the factory.
Bleomycin (Big Bleo) is a hunky guy who everyone admires. Tall and Blonde, hes
also got a pretty nice package (curative for germ cell ovarian and testicular tumors).
Hes also super nice (minimal myelo and immunosuppressive toxicity). Vinblastin
(testicular tumors) wants to work with him and Etoposide (testicular tumor) is
jealous of him (Bleomycin curative of germ cell tumors with vinblastine or
etoposide).
The second group of the DNA intercalating team are the brothers, Dox and Daun.
They are pretty solid dudes (solid tumors). They add diversity to the team as they
are African American. Doxorubicin (Dox) is the bigger brother, and he is also
broader than his little brother Daunorubicin (Dox is more broad spectrum than
Daun). Dox has heart problems (can induce cardiomyopathy) and often has to
pee when you get him red in the face/angry (red urine). Dox often goes to his
physician Dr. Dex, iron chelating specialist, to get his heart checked out
(Dexrazoxane, iron chelator, reduces free radical generation, protects
against cardiomyopathy). Just like the assembly line workers (Vinca twins and
Taxanes), and the janitorial staff (the Posides and the Tecans), the DNA intercalating
team is also under the supervision of the floor manager, Mr. DR, the 1 st (MDR-1).

The next group is the Alkylating Agents, aka the nerds, their job is more technical
than the intercalating team (highly reactive compounds that cause cross
linking of DNA, and also-DNA- protein crosslinks to prevent DNA
replication and transcription of RNA). They are high functioning and not fun to
be around (highly reactive toxic compounds). They are overconfident and can
make the DNA even worse (mutagenic/carcinogenic; can induce secondary
leukemia). They cant stand PJs coffee shop next door to the factory (p53
mutants have intrinsic resistance to alkylating agents). Since they are
specialized, Mr. DR, the 1ss, (MDR-1) the floor manager is NOT their boss.
Nerd classifications:
1. Mustard lovers (Nitrogen mustards): Cyclo and Mel are mustard lovers.
a. Cyclophosphamide (Cyclo) : Broad guy with a beard (broad
spectrum), Burkitts Lymphoma specialist

i. Often gets Hemorrhagic nose bleeds after looking at his Mensa

profile (Hemorrhagic cystitis toxicity, treat with Mesna,
detoxifies urotoxic metabolites).
b. Melphalan (Mel): Mel is a Multiple Myeloma specialist. Hes a pretty
easing going and well-spoken nerd like Cyclo (both well absorbed
orally).
2. Nitro drinking nerds (Nitrosoureas): The mustine brothers Car and Lo,
and their friend Strepto are nitro drinkers.
a. Carmustine and Lomustine: skinny nerds that are pretty smart and
can get in your head (Penetrate CNS well, effective against
astrocytomas and metastatic brain tumors).
b. Streptozotocin (Strepto): middle aged guy who hangs out with the
mustine brothers. He switched over to drinking nitro rather than
alcohol because he has pancreatitis (Metastatic pancreatic islet
cell carcinoma).
3. Water Drinking nerds (Hydrazines): the carbazine brothers Pro and Da
(D.A)
a. Procarbazine and Dacarbazine: Ivy League educated, proper nerds.
Only drink water (hydrazines). Like to read books on Hodgkins
disease. Like working with the well-spoken (orally tolerated)
mustard loving nerds (Cross-resistance with mustard type
alkylating agents).
4. Platinum wearing nerds (Platinum compounds): The platin sisters: Cis
and Carbo platin. Both sisters are a little hard of hearing (ototoxicity).
a. Cisplastin: Cis is also a little sex crazy (testicular cancer):
therefore, her obvious choice of companionship in the factory is with
Bleo, Eto, and Vinblastine.
b. Carboplastin: Carbo likes to hang out with bearded Cyclo and the
Mr. Pac Taxel from the microtubule assembly line (paclitaxel Taxane)
to talk about ovarian cancer.
Besides these blue collar people working in the factory, there are some highly
skilled people that also work there. They are the called RDC team (Rationally
Designed Chemotherapeutics) Team. Their job is to pinpoint and target the
specific things that can go wrong during the cell cycle and signaling pathways.
1. Small group teams ( small molecule inhibitors)
a. The nibs Vemurafenib, Dabrafenib, Trametenib, and Imantinib
i. Vermura and Dabra, the beauty queens and first cousins are in
the skin product line (melanomas, etc).
ii. They are working on inhibiting mutant B-Raf kinase to prevent
Raf (Rouge) skin.
iii. Tramet (Trametenib) works with Vermura and Dabras team
to inhibit mutant B-Raf kinases downstream targets MEK
1 and MEK 2 to make sure people get clear skin.
b. Ima (Imantinib) : Grandma Ima of the nib family is originally from
Philadelphia (Philadelphia chromosome, fusion gene, chr 22)

and is interested in Chronic myelogenous leukemia research

(CML).
i. Grandma Ima is working on inhibiting bcr-abl tyrosine
kinase which causes 95% of all CML. BCR-ABL is a fusion gene
and therefore only in cancerous cells not healthy cells.
