Retroviruses are one of the most efficient tools for delivering genes into dividing mammalian cells. The pCHAC-MCS1-2A-MCS2 Retroviral
Expression Vector is a Moloney Murine Leukemia Virus (MMLV) based vector containing retroviral LTRs and packaging signal. In addition, the
vector also includes two multiple cloning sites (MCS1 and MCS2 on the map below), that can be used to insert two cDNAs to be separated
by 2A peptide sequence. This setup allows bicistronic expression from one transcript with the flexibility of putting different fluorescent protein,
drug resistant gene, or other proteins either as the leading or trailing translation unit.
The 2A-based bicistronic expression has been used for several years, but recently gained much more popularity due to its successful use in
iPSC generation which required 2 to 4 factors working in concert. Even expression of all factors can be achieved when 2A peptides are used
for multiple protein production, due to near 100% efficiency of the 2A “cleavage” at each site, and no interference between multiple 2A sites.
