    Experiment: Starch agar plates


    To prepare in advance by tech: Method
     Starch agar plates Starch agar plates 1. 5g starch in total volume 500mls.
     Barley seeds to germinate (week before) Make starch solution (200mls) 2. Make agar: 14g nutrient
     Yeast solution (48hrs) 25 degree centigrade) agar +teaspoon of agar powder into small amount of water
    heat to about 90c in water bath then cool to 55c. 3.Mix
    solutions and make up 500mls. 4. Autoclave and pour
    plates.
    Items required by class: `
     Plates
     Barley
     Yeast
     Distilled water
     Cork borers
     Dropping pipettes
     Scalpels
     Labels
     Bowl of disinfectant for beakers and pipettes with saliva.
     Cutting tiles
    Notes:
     Plates need to be incubated at 25 c till next lesson.
     Iodine needed next lesson
     Autoclave plates after use.
