Abstract

An animal tissue culture laboratory needs many types of equipment and consumables and the most important thing a sterile environment. We have tried our best to do this by adopting as many as possible aseptic techniques such as using separate sleepers for laboratory, whipping every instrument regularly with 70% ethanol, avoiding eating and talking much in the laboratory etc. Lymphocyte isolation optimization was carried out by isolating lymphocyte under varied condition and was finally observed that lymphocyte yield was highest when undiluted blood was layered over 10% Ficoll and centrifuged at 3300rpm. Chick embryo cells or fibroblasts were a good option for culturing as they can be cultured under less specific and sophisticated condition that is exactly what we can expect in a newly established laboratory. The effect of trypsin and trypsin + versene were observed and can be concluded as more viable cells are obtained when for neutralizing trypsin no specific substance is used (serum or any trypsin inhibitor) but only diluted with medium (RPMI1640). Chick liver show great regenerative power and thus can be hoped to get cultured under less sophisticated condition. Cells were disaggregated with trypsin and cultured in F12 medium.
