Observation:

Preparing and examining slides of plant cell (onion cell) and animal cell (cheek cell)

Material: An onion bulb, iodine solution, distilled water, methylene blue solution.

Apparatus: glass slides, cover slips, light microscope, scalpel, glass dropper, mounting needle, filter paper and toothpick

Procedure: 1. A leaf from a bulb of an onion is peeled off. 2. An epidermal layer from the leaf is peeled with a razor blade. 3. The epidermal layer is then placed on the glass slide that has a drop of distilled water on it. Ensure the epidermal layer does not folded.

A cover slip is placed at an angle over the water droplet on the slide. Slowly slide the cover slip to cover the specimen without trapping any air bubbles. The slide is observed under a light microscope starting with the lowpowered lens, followed by the highpowered lens. A drop of iodine is added to the end of the cover slip and allowed to seep beneath the cover slip.

A strip filter paper is placed at the other end of the cover slip to absorb any excess iodine and water. The slide is observed under the microscope again. The structure of the onion cell is drawn. The experiment is repeated using cheek cell.

The inside of the cheek is scraped with a toothpick. Steps 3-4 is repeated but using cheek extract. A drop of methylene blue solution is placed at one end of the cover slip and a strip of filter paper is placed at the other end of the cover slip.

The slide is then observed under the light microscope. The structure of cheek cell is drawn.

Observation:
Draw the structure that you see under microscope!!

Discussion: 1. Plant cell have a fixed shape. 2. Nucleus, cell membrane, cytoplasm, vacuole and cell wall can be observed under a light microscope. 3. Nucleus give brownish color after stained it with iodine solution. 4. Hydrilla sp. leaf can be use as specimen to observed the plant cell.

Human cheek do not have fixed shape. The cheek cell give blue and have dark blue spot after stained it methylene blue solution. The dark blue spot is a nucleus. Cell membrane, cytoplasm and nucleus that can observed through light microscope.

Conclusion: 1. Plant cells have regular shape. All plant cells contain a nucleus, cell wall, plasma membrane, cytoplasm and a big vacuole. 2. The human cheek cells are not regular in shape. Each human cheek cell contains a nucleus, cytoplasm and plasma membrane.
