Differentiation Citrate+/Citrate1. Preparation of the medium Base A: Tryptone 20g Yeast extract 5g Gelatin 2.

5g Glucose 5g Lactose 5g NaCl 4g Trisodium Citrate 2g Calcium Lactate 8g Agar 15g Distilled Water 1000ml Verify pH: 6.6/6.7 Sterilize for 15 minutes at 120C. Let cool to 50C. Base B: Serum: Obtain by paper filtration of a fresh starter and then sterilize for 15 minutes at 120C. Base C: Suspension of Calcium Citrate : Take Calcium Citrate 4H2O. Suspend 10g of Citrate in 100ml of 1.5% solution of Carboxymethylcellulose at 45-50C. Mix everything together. Sterilize for 15 minutes at 120C. For 100ml of Base A, add 5ml of Serum and 5ml of Calcium Citrate. The medium is used in population counting. Incubate for 3 days at 25C.

Legend: Citrate+ : Lactococcus lactis subsp. lactis biovar diacetylactis Leuconostoc mesenteroides Presence of a transparent halo around the colony.

Citrate+ : Lactococcus lactis subsp. lactis Leuconostoc mesenteroides subsp. Cremoris No halo.
