    Warm up spectrophotometer according to protocol. Blank spectrophotometer with 0.8-1 mL of culture media in a cuvette (ie. LB). Pipette 0.8-1 mL sample of your culture into cuvette and take reading. Sample can then be diluted appropriately using the following formula. o Desired OD/Sample OD = % of sample in final volume

    Retest OD after mixing sample thoroughly (swirling or briefly vortexing) to attain new OD.
