Instructions for Use
Preparation
Warm TS vial with a cap and a Petri Dish in an incubator to 37C.Pour
the full content of TS into the Petri Dish.
Drop 300l each for DS, WS1 and WS2 on the Repro Plate with micro
pipette.
NOTE: Use pasteur pipette that has a suitable internal diameter for
Oocyte(External diameter:140m) or Embryo (140~180m).
Thawing
1. Quickly immerse Cryotop sheet into TS. Leave it for 1 minute.
2. Aspirate the Oocyte (Embryo) with the pasteur pipette and gently place it
on the BOTTOM of DS. Leave it for 3 minutes.
3. Aspirate the Oocyte (Embryo) with the pasteur pipette and gently place it
on the BOTTOM of WS1. Leave it for 5 minutes.
4. Aspirate the Oocyte (Embryo) with the pasteur pipette and gently place it
on the TOP of WS2. After the Oocyte (Embryo) free-falls to the bottom of
WS2, do the same work twice in WS2.
5. Transfer the Oocyte (Embryo) to a culture dish containing the
appropriate culture medium. Incubate the Oocyte (Embryo) in a 37C
incubator to complete recovery.
NOTE: Oocytes for 2 hours and Embryos for 3 hours as an idea.

Quality Control Testing

Each lot of Vitrification Kit-Thawing Media receives the following tests:

Mouse Embryo Assay (One Cell)

Preparazione
Riscaldare la fiala di SS con un coperchio e la piastra di Pietri in
unincubatrice a 37. Versare tutto il contenuto di SS nella piastra di Pietri.
Versare 300l di SD, SR1 e SR2 sulla piastra Repro con la micro pipetta.
Nota: Utilizzare una pipetta di Pasteur che abbia un diametro interno
adeguato allovocita (diametro esterno: 140m) o allembrione
(140~180m).
Scongelamento
1. Immergere velocemente il foglio di Cryotop in SS. Lasciarglielo un
minuto.
2. Aspirare lovocito (embrione) con la pipetta di Pasteur e posarlo
delicatamente sul FONDO di SD. Lasciare 3 minuti.
3. Aspirare lovocito (embrione) con la pipetta di Pasteur e collocarlo
delicatamente sul FONDO di SR1. Lasciarlo 5 minuti.
4. Aspirare lovocito (embrione) con la pipetta di Pasteur e collocarlo
delicatamente IN CIMA a SR2. Dopo che lovocito (embrione) scende IN
FONDO a SR2, ripetere due volte in SR2.
5. Trasferire lovocito (embrione) in una piastra di coltura contenente il
mezzo di coltura appropriato. Incubare lovocito (embrione) in
unincubatrice a 37 per completare la recovery.
Nota:Ovociti per 2 ore ed embrioni per 3 ore.

Test di controllo di qualit

Ogni confezione di Kit Cryotop per lo scongelamento viene sottoposta ai
seguenti test:

Endotossina tramite Lal test (con utilizzo di topi)

Gebrauchsanweisungen

Prparation
Rchauffez 37C la fiole de la solution de dconglation (TS) avec un
couvercle et une bote de Ptri dans l'tuve culture. Versez tout le
contenu de la solution de dconglation (TS) dans la bote de Ptri.
Versez 300l de chacune des solutions de DS, WS1 et WS2 sur la plaque
Repro l'aide de la micro-pipette.
NOTE: Veillez utiliser une pipette Pasteur dont le diamtre interne soit
adquat pour les Ovocytes (Diamtre externe: 140m) ou pour les
Embryons (140 180m).

Vorbereitung
TS Ampulle mit Deckel und die Petrischale in einem Wrmekasten
(Inkubator) auf 370C erwrmen. Den gesamten Inhalt der TS Lsung in die
Petrischale gieen.
Fllen Sie jeweils 300l mit einer Mikropipette fr die DS, WS1 und WS2
Lsungen in die Reproschale.
ACHTUNG: Benutzen Sie Pasteur Pipetten, die einen angemessenen
inneren Durchmesser fr Oozyten (uerer Durchmesser 140 m) oder fr
Embryos (140 180 m) besitzen.

Dconglation
1. Plongez rapidement la feuille Cryotop dans la solution de dconglation
(TS). Laisser agir 1 minute.
2. Aspirez l'Ovocyte (Embryon) avec la pointe de la pipette et placez-le
dlicatement sur la BASE de la DS. Laissez agir 3 minutes.
3. Aspirez l'Ovocyte (Embryon) avec la pointe de la pipette et placez-le
dlicatement sur la BASE de la WS1. Laisser agir 5 minutes.
4. Aspirez l'Ovocyte (Embryon) avec la pointe de la pipette et placez-le
dlicatement sur la PARTIE SUPRIEURE de la WS1. Une fois que
l'Ovocyte (Embryon) tombe librement dans la base de la WS2, refaites cette
opration une deuxime fois sur la WS2.
5. Transfrez l'Ovocyte (Embryon) une plaque de culture qui contient le
milieu de culture appropri. tuvez l'Ovocyte (Embryon) dans l'tuve
culture 37C jusqu'' ce qu'il soit compltement rcupr.
NOTE: les Ovocytes pendant 2 heures et les Embryons pendant 3 heures.

Auftauen
1. Tauchen Sie das Cryotop Blatt schnell in die TS Lsung ein und lassen
Sie es dort 1 Minute lang.
2. Heben Sie den Oozyten (Embryo) mit der Pasteur Pipette heraus und
legen Sie ihn dann vorsichtig auf den Boden der DS Lsung. Lassen Sie
ihn dort 3 Minuten lang.
3. Heben Sie den Oozyten (Embryo) mit der Pasteur Pipette heraus und
legen Sie ihn dann vorsichtig auf den Boden der WS1 Lsung. Lassen Sie
ihn dort 5 Minuten lang.
4. Heben Sie den Oozyten (Embryo) mit der Pasteur Pipette heraus und
befrdern Sie ihn dann vorsichtig auf die Oberflche der WS2 Lsung.
Wenn der Oozyt (Embryo) von selbst auf den Boden der WS2 Lsung
gefallen ist, wiederholen Sie diesen Vorgang noch 2 Mal in derselben
Lsung, WS2.
5. Befrdern Sie den Oozyten (Embryo) in eine Kulturschale, die einen
angemessenen Nhrboden enthlt, lassen Sie ihn dann in einem Inkubator
bei 370C bebrten, um ihn wieder vllig herzustellen.
ACHTUNG: Oozyten 2 Stunden lang bebrten lassen, Embryos 3 Stunden
lang.

Odmraanie
1. Szybko zanurzy arkusz Kriotopu w roztworze TS. Pozostawi na 1
minut.
2. Zassa komrk jajow (zarodek) przy pomocy pipety Pasteura i
delikatnie umieci na DNIE DS. Pozostawi na 3 minuty.
3. Zassa komrk jajow (zarodek) przy pomocy pipety Pasteura i
delikatnie umieci na DNIE WS1. Pozostawi na 5 minut.
4. Zassa komrk jajow (zarodek) za pomoc pipety Pasteura i delikatnie
umieci na POWIERZCHNI WS2. Kiedy komrka jajowa (zarodek)
opadnie swobodnie na dno WS2, powtrzy t sam czynno dwukrotnie
w WS2.
5. Przenie komrk jajow (zarodek) do naczynia zawierajcego
odpowiedni poywk do hodowli. Inkubowa komrk jajow (zarodek) w
inkubatorze w temperaturze 37 C, w celu zakoczenia procesu.

Storage Temperature

Uso previsto
Cryotop safety Kit Descongelacin se usa para descongelar Ovocitos y
Embriones humanos vitrificados.

Cryotop safety Kit Descongelacin

N 1. Solucin de descongelacin (TS): 2 vial X 4.0 ml
N 2. Solucin diluyente (DS): 1 vial X 1,5 ml
N 3. Solucin de Lavado (WS): 1 vial X 1.5 ml
2 Placa de Petri: 35mm para la solucin de descongelacin
1 Placa Repro: con 6 senos

Instrucciones de Uso
Preparacin
Calentar el vial de la solucin de descongelacin (TS) con una tapa y una
placa de Petri en una incubadora a 37C. Verter todo el contenido de la
solucin de descongelacin (TS) en la placa de Petri.
Verter 300l de cada una de las soluciones de DS, WS1 y WS2 en la
placa Repro con la micropipeta.
NOTA: Utilizar una pipeta Pasteur que tenga el dimetro interno adecuado
para los Ovocitos (Dimetro externo: 140m ) o los Embriones (140
180m).
Descongelacin
1. Sumergir rpidamente la hoja Cryotop en la solucin de descongelacin
(TS). Dejarla 1 minuto.
2. Aspirar el Ovocito (Embrin) con la pipeta Pasteur y colocarlo
suavemente sobre la BASE de la DS. Dejarlo 3 minutos.
3. Aspirar el Ovocito (Embrin) con la pipeta Pasteur y colocarlo
suavemente sobre la BASE de la WS1. Dejarlo 5 minutos.
4. Aspirar el Ovocito (Embrin) con la pipeta Pasteur y colocarlo
suavemente en la PARTE SUPERIOR de la WS1. Despus de que el
Ovocito (Embrin) caiga libremente en la base de la WS2, repetir esa tarea
una segunda vez sobre la WS2.
5. Transferir el Ovocito (Embrin) a una placa de cultivo que contenga el
medio de cultivo adecuado. Incubar el Ovocito (Embrin) en una
incubadora a 37C hasta que se recupere completamente.
NOTA: los Ovocitos durante 2 horas y los Embriones durante 3 horas.

Pruebas de Control de Calidad

A cada lote de los Cryotop Safety Kit Descongelacin se le realizan las
siguientes pruebas:
Soluciones:
Esterilidad mediante Test de esterilidad UPS (SAL 10-3).
Endotoxinas mediante la metodologa LAS en ratones.
