Zika
The Zika virus outbreak is now considered an international public health
emergency by the World Health Organization, a rare move that illustrates the
severity of Zika and the desire to encourage further research and tackle some of
the biggest unanswered questions about the virus.
1. Does Zika cause microcephaly?
As Zika gained the media spotlight over the past few weeks, there are still many
unknowns about the mosquito-borne virus. The most well-documented,
although yet unconfirmed, aspect of Zika is its association with microcephaly
brain damage in infants born to infected mothers. Brazil has reported nearly
4,000 suspected cases of microcephaly, in which infants were born with smallerthan-usual brains.
"The Brazilians believe there is a strong connection with microcephaly that we
are seeing in their population," said Dr. Ron Warner, director of the Texas Tech
Physicians Travel Medicine Clinic. "I suspect it will be several weeks before
they are able to be more definitive about this link."
Adding a layer of speculation to this possible connection, Colombia, which has
the second largest number of confirmed cases of Zika after Brazil, reported no
cases of microcephaly in its population of more than 2,100 pregnant women
infected with the virus, Dr. Warner added.
2. What prompted the outbreak?
Another major question is what started the Zika outbreak. The first reported
cases of Zika in humans occurred in 1947 in Uganda, though the exact reason
for the virus' proliferation today has not been specified. While some point to an

infected traveler spreading the virus to other attendees of the 2014 World Cup in
Brazil, others note that the outbreak of Zika is related to chance rather than a
certain event.
"Why the virus has spread now as opposed to not before is largely a matter of
chance," said Alex Perkins, a biological science professor at Notre Dame
University. "It is likely the wrong person at the wrong time happened to be in
Brazil to start a local outbreak that we see today."
Recently, a claim that the virus was supposedly spread through genetically
modified mosquitoes notes the Zika outbreak epicenter is the same location
where mutant mosquitoes were released in 2015. Blogs circulated this story
criticizing the biotech firm Oxitec that headed the experiment by releasing GM
mosquitoes in Brazil to compete with normal Aedes aegypti mosquitoes that
transmit the virus.
Oxitec vehemently denied this claim, explaining that the first cases in Brazil
were reported in May 2015 in the city of Camaari in Bahia State, where no
trials have taken place. The trial sites are therefore over 400 kilometers (about
249 miles) from the centers of disease outbreak, which is significantly more
than the 200-yard distance the mosquitoes can travel in their lifetime.
"Even if an Oxitec mosquito 'hitched a lift,' it would die and not be able to
establish a population," said Oxitec CEO Hadyn Parry.
The link between the release of genetically modified mosquitoes and Zika is
also "complete nonsense" because Oxitec releases male mosquitoes that cannot
transmit the disease as they do not bite, added Parry.
"I think this connection is completely implausible for all sorts of reasons," said
Perkins. "It is unfortunate that this conspiracy is spread because it has negative
health consequences as these mosquitoes can get rid of the Aedes."
As the exact origin of the virus is unclear, blaming a type of technology or a
single event can provide an easy explanation for Zika's global presence.
3. How bad will a Zika outbreak be in the U.S.?
Forward-looking questions about Zika's spread are also top of mind within the
scientific community. Thus far, local transmission of the Zika virus has not been
identified in the continental U.S., so a common question being asked of the
research community is how severe the outbreak of the virus might be if it is
transmitted to the states.
"I don't think Zika will be well-established in the U.S.," said Dr. Anna Durbin,
an associate professor at the Johns Hopkins Bloomberg School of Public Health.
"We will probably see outbreaks in areas like the Florida Keys and the Gulf
Coast. The infrastructure we have in most of our buildings will prevent the virus
from establishing itself in the same way that it has in Brazil."
Though this mosquito is typically found in tropical locations, the mainland U.S.
does have Aedes species mosquitoes that can become infected with and spread
Zika virus, according to the CDC. The virus is transmitted when an Aedes

mosquito bites a person with an active infection and then spreads the virus by
biting other individuals. During the time when those people have symptoms,
they can then become carriers. Transmission in the U.S. may start to happen as
the weather becomes warmer, however, the propagation of the virus is less
predictable.
"As much attention as Zika is getting in the media, it is still difficult to ascertain
what is really going on and exactly how bad the problem is," said Perkins. "The
ongoing nature of this threat and the limited capability we have to diagnose and
confirm 80 percent or more of Zika infections pose challenges to studying it and
responding to it, but this WHO declaration is a step in the right direction."

Concerns have been raised scientists could be to blame for the Zika virus
outbreak after genetically modified insects were released into Brazil three
years ago.
Didcot-based biotechnology company Oxitec worked on the same type of
mosquito that carries the virus in 2012, engineering them to have offspring that
die out before they can breed, reducing the population of disease-carrying bugs.
Some scientists questioned the wisdom of the plan at the time and it has led to
critics claiming the modification could have sparked the current outbreak.

Before a virus can be said to cause disease, a few procedures need to occur.
First, the virus must be proved to exist. It has to be isolated from a human
carrier as diseased tissue, and then that tissue has to be put under an electron
microscope, where many, many (Zika) viruses can be seen. Second, tests have
to be run on many suspected human cases, and these tests have to reveal very
large amounts of Zika in the body. Thats your basic starter kit for deciding that
a virus might be causing actual human disease.

Synthetic biology a eugenic philosophy.

"Synthetic biology" is an umbrella term that refers to a new set of powerful
techniques for manipulating the fundamental molecular structures of life,
including genes, genomes, cells and proteins. Techniques being developed
under the "synthetic biology" rubric include the modification of existing
bacteria to produce useful substances or perform new functions, the creation of
novel artificial organisms from "scratch," and less noted to date the
modification of animal and human genes. Synthetic biologists foresee a host of
human applications, including new methods to produce drugs, biofuels and
vaccines; to diagnose, prevent and cure disease; and far more controversially
to screen, select, and modify genes for specified traits in embryos, children,
and adults. Nonetheless, the field remains in its early days, and separating
hype from real potential remains difficult. While diverse constituencies have
voiced concerns about ecological and biosecurity risks, little attention has so far
been called to the dangers connected to synthetic biology's human applications.
Synthetically engineered viruses and pathogens and synthetic organisms
released in the human body such as "tumor eating" bacteria, for example, pose
profound dangers to human health. Synthetic biology also presents dangers of a
different kind if the field spawns forms of human genetic manipulation that
heretofore have been impracticable. These include human reproductive cloning,
the creation of "designer babies" through inheritable genetic modification, and
other purported "enhancements." Leading figures in the synthetic biology field
have in fact predicted, and in some cases embraced, such eugenic visions. Such
prospects raise concerns for social justice, human rights, and equality. However,
at present, no comprehensive framework for assessment, oversight and
regulation of synthetic biology exists nationally or internationally.

fairly small company, and decides to turn it into a big one without using venture
capital. His goal is to make it "the Google of the life sciences." Among the
acquisitions and partnerships he makes are: a company developing personalized
cell and gene therapies using iPS cells an animal cloning company (both the
agricultural and pet markets) a biopharmaceutical company focused on cancer
immunotherapies a drug development and delivery company a company that
makes genetically modified fish (even before selling them was legal) a company
that makes sterile mosquitoes in order to effect permanent germline changes a
company that makes genetically modified apples a company pushing hard into
multiple IVF markets around the world, with products not currently legal in the

U.S., and research into human egg precursors with the explicit intention of
producing hundreds of eggs and possibly embryos and the ambition of
editing them In short, lots of private money, deep connections with the
pharmaceutical industry, a major focus on synthetic biology, a willingness to
jump borders for legal convenience, a deep interest in reproductive
technologies, and the clear intention to work on "enhancing" embryos. That
could turn into a nightmare. But it's real, and it's happening now.

The billionaire is Randal Kirk, the main company Intrexon, which cannily
snagged the URL dna.com. The iPS company is Fibrocell; the cloning one
isViagen (part of Trans Ova Genetics); the (disappointing so far)
biopharmaceutical is Ziopharm; the drug company is Halozyme; the fish
areAquAdvantage salmon made by AquaBounty; Oxitec makes the sterile
mosquitoes; the Arctic Apple was developed by Okanagan; and the IVF
company is the extremely controversial OvaScience.
Kirk, a lawyer by training, made his first fortune ($65 million) with a medical
supplies company; his first big one with New River Pharmaceuticals (he
cleared $1.2 billion); his second with Clinical Data (roughly $600 million); and
his third when he took Intrexon public in 2013 (his shares have been valued
at $1.5 billion). He avoids outside venture capital, and held onto over 60% of
Intrexon; the success of its IPO may have been largely because of his existing
reputation.
Clearly Kirk bought Intrexon as a way into synthetic biology. Its founder,
molecular geneticist Thomas Reed, positioned what was originally called
Genomatix as a gene-tools or DNA-parts venture, but Kirk had much broader
ambitions (yes, the Google quote was his, though he said it before Google

became the Google of the life sciences). In a 2011 profile, Forbes also reported
Kirk's prediction that:
in a decade it [Intrexon] could become "the largest, most significant company"
in its burgeoning field.
He could be right.
Of course, he also might not. Indeed, Viagen was a spin-off from Genetic
Savings and Clone (GSC), a pet cloning company that was set up by the
billionaire John Sperling. GSC was finally abandoned, partly for technical and
ethical reasons and above all because it was not commercially viable.
Intrexon and OvaScience
Last April, MIT Technology Review noted that OvaScience intended to "correct
[harmful genetic] mutations before we generate your child," and Motley
Foolpointed out the potential synergies between OvaScience and Intrexon,
focused on a joint venture called OvaXon, described on the Intrexon website
thus:
The joint venture looks to create new applications for improving human and
animal health. Under the joint venture, OvaScience's EggPC platform will be
combined with Intrexon's genome engineering capabilities with the goal of
offering an innovative approach for the prevention of inherited diseases in
humans, such as mitochondrial and other genetic disorders.
The technology behind OvaScience is scientifically controversial, ultimately
based on the disputed discovery of "egg precursor cells" that "have the potential
to develop into mature eggs, thereby replenishing a womans egg supply." Its
main product at present is Augment, which "uses the energy-producing
mitochondria from your own egg precursor (EggPC) cells to supplement
the existing mitochondria in your eggs" and thus supposedly improve fertility.
One of the founders of OvaScience told a stock analyst last spring that
usingCRISPR in germline engineering was not on the current agenda ("they
have enough headaches at this point") but they do admit to being aware of
thepotential to use it that way.
The FDA will not allow Augment to be sold in the U.S. without clinical trials.
However, it is sold in Canada, where the first baby was born after that treatment
in May, 2015; and also in Japan, Turkey and Dubai. A British IVF clinic
is applying for permission to use the procedure "in a pilot trial involving about
20 women."
Permission may not be granted in the UK Robin Lovell-Badge, for one, is
extremely skeptical but the attempt illustrates the difficulty of establishing,
let alone enforcing, international standards. The company is trying topressure
the FDA by creating consumer demand here with promotional stories from
abroad.

Intrexon and its subsidiaries have other experience with transnational evasion
techniques: AquaBounty was an American company, founded in Maynard, MA,
and incorporated in Delaware, that set up in Canada to create eggs that would be
grown in Panama. It nearly went bust until Kirk bought in.
Kirk is arguably "Biotech's Best Investor." He's clearly a technophile and
definitely thinks that synthetic biology is the future he's called his partner
Reed "the Henry Ford of DNA" but he does not seem to be primarily an
ideologue; he wants to make another boatload of money.

The World Health Organization announced it will convene an
Emergency Committee under International Health Regulations
on Monday, February 1, concerning the Zika virus explosive
spread throughout the Americas. The virus reportedly has the

Zika seemingly exploded out of nowhere. Though it was first
discovered in 1947, cases only sporadically occurred
throughout Africa and southern Asia. In 2007, the first case was
reported in the Pacific. In 2013, a smattering of small outbreaks
and individual cases were officially documented in Africa and
the western Pacific. They also began showing up in the
Americas. In May 2015, Brazil reported its first case of Zika
virus and the situation changed dramatically.
