Abstract
TrichodermaharzianumwasisolatedfromlocaldumpsitesofShivamoggaDistrictforuseinthebiodegradationofpolyethylene.Soilsampleof
thatdumpsitewasusedforisolationofT.harzianum.Degradationwascarriedoutusingautoclaved,UVtreated,andsurfacesterilized
polyethylene.Degradationwasmonitoredbyobservingweightlossandchangesinphysicalstructurebyscanningelectronmicroscopy,Fourier
transforminfraredspectroscopy,andnuclearmagneticresonancespectroscopy.T.harzianumwasabletodegradetreatedpolyethylene(40%)
moreefficientlythanautoclaved(23%)andsurfacesterilizedpolyethylene(13%).Enzymesresponsibleforpolyethylenedegradationwere
screenedfromT.harzianumandwereidentifiedaslaccaseandmanganeseperoxidase.Theseenzymeswereproducedinlargeamount,and
theiractivitywascalculatedusingspectrophotometricmethodandcrudeextractionofenzymeswascarriedout.Molecularweightoflaccase
wasdeterminedas88kDaandthatofmanganeseperoxidasewas55kDa.Thecapacityofcrudeenzymestodegradepolyethylenewasalso
determined.Byobservingtheseresults,wecanconcludethatthisorganismmayactassolutionfortheproblemcausedbypolyethylenein
nature.
