Precautions for Safe Operation


The RID-1OA is the differential refractive index detector for the high performance liqid
chromatograph.
In order to operate the unit for safety, strictly observe the following points.
1.

Do not use the unit for the purpose other than the above mentioned analysis.

2.

Follow the procedures described in the instruction manual.

3.

Observe the warnings and cautions.

4. Do not disassemble or modify the unit without our approval. Failing to do so may lead
dangerous situation or damage of the unit.

NOTE
This instruction stipulates the content of warnings as follows:

Applied for the case where death or serious injury is assumed


to happen at any moment to the repair engineer when a mistake
is made in the work or to the user due to the nonconformity of a
product occurring after the work.

When operating the parts where this mark is indicated, exercise special caution after reading

Precaution on installation site and handling- of the unit

Generally. a large amount of organic solvents is used in the high-performance liquid


chromatograph. Sufficient care should be taken in installation site and handling of the unit.
Please take care of the notices in the text, not to mention the precautionary requirements listed
below :
1.

Vent ilat ion


Solvents used in the high-performance liquid chromatograph are inflammable or toxic. Be
sure to ventilate the room well.

2.

Fire
Open flames are prohibited in the room where high-performance liquid chromatograph is
installed. Avoid installing units which emit sparks in the room.
It is also necessary to provide a fire extinguisher preparing for the worst.

3. Safety goggles
Wear safety goggles in handling solvents.
4.

Other equipment
A washstand or the like is necessary to be furnished nearby preparing for the cases such that
solvent comes into the personnel's eyes or the personnel touches toxic solvent.

5.

Power source
Working voltage range and power consumption of this system are as follows.
Be sure to connect to the power source conforming to them.

To prevent electric shock and to secure safe operation of the system, always make
grounding.

Repair and maintenance of the unit

Normal maintenance of this unit can be performed without removing the cover. Do not
remove the cover at the normal maintenance. Contact our sales office or agent of repair
requires to remove the cover of the main body.

Solvent not usable


The following solvents affect the materials used in the wetted part and deteriorate the
strength of the material. In the worst case, pipes may burst and high-pressure solvent
scatters.
HFIPA (Hexafluoroisopropyl alcohol)
Others
The equipment should not be exposed to direct sunlight or strong air currents. It is also
recommended to install in a room where temperature fluctuation is small.

Precautions on Static Electricity


Liquid chromatography using flammable organic solvents as mobile phase requires proper care
against fire, explosion, etc. Particularly, among various possible accidents, those caused by static
electricity are difficult to anticipate, and tend to occur only with unexpected conditions which
often make countermeasures insufficient.
At a site where preparative liquid chromatography is practiced, a large amount of flammable
substances may be used. Therefore, once an accident happens, it could lead to tremendous
damage.
The mechanism of accident caused by static electrical discharge and preventive measures are
described below. Take due care in safety measures in handling of equipment.

Mechanism of Static Electrical Discharge Accident (Example)


Accidents caused by static electricity take place through the following processes.

When liquid is fed at high speed through a small-diameter


tube like the pipe of a liquid chromatograph, static electrical
charge occurs by friction between solid and liquid as shown in
Fig. 1.

Charging and storage
of static electricity

When the charged liquid is collected in an insulated vessel,

the static charge accumulates gradually, and the voltage can


easily reach a few kilovolts.

Preventive Measures against Accidents


The principal preventive measure is the prevention of charging and storage of static
electricity among those items shown in Occurrence mechanism of static electrical
discharge accidents. The preventive measures are shown below. It is recommended to
exercise two or more measures simultaneously.
+E Particularly when a large quantity of flammable solvent is held in a large vessel, be sure
to observe the preventive measures 1,2, and 3.

Preventive measure 1.
Use metallic (conductive) waste liquid vessel which is well grounded. This releases the
charge of the waste liquid and vessel to ground.
The following items are available.
(1) Grounding wire with clip
P/N 228-21 353-9 1
(2) Metallic 18 liter can

(3) Metallic 4 liter can

% Be sure to ground the vessel properly. Disconnecting of grounding wire or poor
grounding defeat the purpose of using a metallic vessel.
There are some metallic cans which have no conductivity due to an oxidized coating
or lacquer on their surface. Be sure to confirm the grounding of vessels by a tester
before application.
% When a liquid with almost no conductivity (of 10-%./mor less) is discharged into the
vessel, it is necessary to mix it with another liquid with some conductivity. (The other
liquid can be placed in the vessel in advance.)

Minimize the clearance of both inlet and outlet of vessel to prevent flame from entering the
vessel.
(1) Cap with three holes for 18 liter and 4 liter cans (P/N 228-21354-91) is available.

Preventive measure 3.
Do not approach the vessel with charged objects including the human body.
Charging prevention measures for human body
Prevention of charging of shoes and clothes
Grounding of human body
Make working floor conductive
Suitable products to be used for those measures a), b), and c) are available on the
market.
When persons who use no charge prevention measures approach dangerous sections,
they have to be grounded beforehand. (For example, they should contact grounded
metal by hand.)
Preventive measure 4.
Use pipes with inner diameter of 2mm or more for waste liquid line for large flow rates.
2 Inclusion of bubbles in the tube may increaqe the amount of charging by ten times.
Check that there is no inclusion of air via tube joints.

Preventive measure 5.
When it is impossible to use a conductive vessel, use caution in the following points.
a) Set the vessel so that the pipe outlet will be placed below the liquid level in the
vessel. Or, dip a grounded metal (ex. pipe connected to the main body of device) in
the liquid.
::: This method is not effective for liquid with small conductivity (lO-los/mor less).
b) Use a vessel of the smallest possible capacity to minimize the damage by fire if it
should occur.
c) Prevent the room from being dry. Humidity of 65% or more has charge prevention
effects.

Introduction

Thls service manual describes maintenance of the IUD-IOA Differential


Refractive Index Detector for S h a d z u High Performance Liquid
Chromatograph. Description of the operating principle, unit configuration, and control circuits as well as how to perform the functions
and performance checks are summed up and edited so that it is easy
to use for a service engineer.
Although some items overlap with those of the instruction manual,
for installation and operation, read the instruction manual along
with this manual as required and perform adequate and effective
maintenance. Also, care should be taken to observe the precautions
for safety which is described in later chapter to avoid any accident.

manual may be partly different from the ones actually used. and

also, we do not stock all the parts described on the disassembly

drawing and/or parts list as repair parts.

Principle of measurement
The following illustration shows the optical system of this unit.
The light radiated from the lamp passes through the lens slit and
passes through the cell in the form of parallel rays, which is, then,
reflected by the mirror and passes through the cell to finally form a
image of the slit on the photodiode.
The flow cell consists of the sample side and the reference side.
When the refractive index in the sample side cell varies, imaze of
the slit travels horizontally.

This unit has a photodiode divided into four as shown in the figure
below. By selecting a combination of photodiode elements, the unit
can provide measurements from high-sensitivity analysis to high
concentration preparative analysis.

Signal processing in mode A

A s i p a l is processed regarding each of the right and the left of the


photodiode as an individual element. Data processing is performed
regarding it as a two-division cell divided into right and left parts.
When the refractive index in the flowcell varies, balance of the
intensity of light into right and left cells changes. The change in the

Signal processing is performed regarding each of the upper and the


lower parts of the photodiode as an individual element. Since the

Same as the mode P, signal processing is performed regarding each
of the upper and the lower parts of the diode as an individual cell.
However, since the reference side with less flow line resistance is
used as the measuring cell, when the refractive index varies, the
image moves in the reverse direction of that of mode P. Therefore
polarity is turned over before change in the balance is converted
into refractive index.

When the flow selection block option is equipped, refer to 5.3.
At measurement
When R.FLOW switch is OFF, the solenoid valve is open at side A.
The liquid from the sample cell does not flow into the referenceside cell, but into the outlet port. Liquid inside the reference-side
cell is sealed in.

When R.FLOW switch is Oh', the solenoid valve is open at side B.


The liquid from the sample flows into the outlet port after passing
through the reference-side cell. This is used to fill the sample cell
and the reference cell with the solvent of equal refractive index

It is necessary with the RID-1OA to select the switch on the switching power supply ECX-OOSD (Section 4.4) when changing the
power supply voltage.
Be sure to disconnect the power supply plug socket before performing the following operations.

Cleaning the Flow Lines

Dirty cells and pipings cause the phenomena such as unstable base
line and large base line noise due to pulsation of liquid delivery
pump. In this case, clean the flow lines with the following procedures.

Mixing nitric acid with organic solvents may form explosive


substance. Be fully careful for handling the waste liquid.
Mount the adapter on the syringe and supply acetone from
the inlet port to clean the flow lines. Press (ShlftandlRflow]
to clean the reference-side flow lines as well.
(2) Supply distilled water and clean the flow lines with the similar procedure.
( 3 ) Next, clean with 0.1 N nitric acid.
(4) Rinse off the dirt and the nimc acid with the distilled water
sufficiently.
( 5 ) Purge the flowline with the mobile phase.
When the mobile phase used for analysis is not soluble with
water, replace the flow lines with acetone and return to the
mobile phase.

The service life of a lamp is 20,000 hours. When the lamp

Mounting the flow selection block

Mounting the optional flow selection block enables selection of the


measurement mode which comprises with each usage of high-sensitivity analysis, semi-preparative, and large-prep.
(With the standard specifications, only the high-sensitivity mode
and semi-preparative mode are available.)

I Note

Mounting of the flow cell block option should be performed by a


service engineer.

9.1 .ICauses of the troubles with liquid and flow lines


Phenomena of the drift, noise, and fluctuation on the baseline are
listed on the below table.
In addition, the following cares should be taken as general.

When the unit is operated under incorrect conditions of carrier

liquid, temperature, and pH, the fixed phase of the column filling map be flown out. This may cause noise.
When solubility of the sample substance to the carrier liquid is

smaller than that to the sample solvent, the sample may be separated out. This may cause noise increase.
When the column is dirty with the sample substance due to

repeated sample introduction, noise and drift may be caused.


Other common causes of drift and noise are slight leak of liquid
from the connection portions and generation of the bubbles
caused due to insufficient degassing. This kind of troubles can
be avoided through sufficient inspection and solution.
