Diffusion of solutes in water

Place 4 test tubes containing 10mlf


distilled water, tap water, hot
distilled water and cold distilled
water.

Place a dot of KMnO4 to each test


tube

Note of the time at which KMnO4


were completely dissolve in water.

Diffusion in a colloid
Melt the agar/ gelatin according to
manufacturers procedure and pour
10ml of the melted gelatin in 8-10
test tubes.

Put 0.5 ml of KMnO4 in one of the


gelatin-tube after it solidifies. Add
the same amount of congo red and
methylene blue in separate tube.

Measure the migration of the dye


starting at the top level of the gelatin
at 10 minutes interval for 60
minutes.

Osmosis (Longanisa skin casing model)

Put 10ml of saturated sugar solution


in a test tube.

Cover the mouth of the test tube


with one ply of longanisa casing and
secure it with a rubber band and
measure the weight of the covered
test tube using a triple beam balance.

Using the Iron clamp, fasten and


suspend the test tube upside down in
a 250-ml beaker containing distilled
water.

Measure the weight of the test tube


for 60 minutes at 10 minutes interval
then tabulate the results.

Osmosis in RBC

Prepare 3 clean glass slides and


cover slips.

Make a puncture at the donors


finger then place a drop of blood in
each glass slides.

Apply a drop of Isotonic-Normal


Saline Solution (0.85 M NaCl) in
first glass slide, Hypertonic Salt
Solution (3.00 M NaCL) to second
g;lass slide and Hypotonic Salt
Solution (0.30 M NaCl) to the
remaining glass slides.

Under the microscope at 40xobjective observe the RBCs. Take


pictures and compare the sizes of the
RBCs of the different slides.

Dialysis

Place a 3ml Methylene blue solution


and 5 ml Saturated CaCl2 solution
in a test tube then cover the mouth
of the test tube with 1 ply of
Longanisa casing and secure the
cover with a rubber band.

Fasten the test tube using an iron


clamp and suspend the test tube
upside down in a beaker (250 mL)
containing distilled water .

Test the presence of Methylene


Blue, Chloride and Calcium in the
beaker-water after every 2 minutes
for 30 minutes.

The presence of Chloride is


identified by adding a drop of silver
nitrate/ Silver sulphate test solution
to 1 ml aliquot of the beakerwater.While the presence of Calcium
is identified through adding a drop
of oxalic acid test solution to ! ml
aliquot of the beaker-water.

Note the results of which molecules


passed through the longanisa skin
first.
