Design,Synthesis, Pharmacological Testing and QSAR Study of 2,4thiazolidinedione as


Antidiabetic Agents.

Abstract:
Peroxisome proliferator activated receptor-gamma (PPARγ) is transducer proteins
belonging to nuclear receptor super family. Interest has been focused on PPARγ because it plays
an important role in type two diabetes mellitus. It has been reported that 2, 4-thiazolidinediones
causes activation of PPARγ receptor. Based on these findings a series of N-substituted 2, 4-
thiazolidinediones derivatives were synthesized and screened for antidiabetic activity.
Ten N-substituted 2, 4-thiazolidinediones derivatives were synthesized by reacting
thiourea with chloroacetic acid to obtain 2, 4-Thiazolidinedione which was then condensed with
substituted benzyl halide to get the title compounds. The structures of finally synthesized
compounds were confirmed by chromatographic and spectral analysis.
All the compounds were screened for antidiabetic activity using alloxan induced diabetes
mice model. 3-Benzyl-2,4-thiazolidinedione and 3-(4-methoxy-benzyl)-2,4-thiazolidinedione
showed hypoglycemic activity comparable to Pioglitazone. Other compounds showed moderate
activity.
QSAR analysis of these N-substituted 2, 4-thiazolidinediones was performed using
multiple linear regression (MLR) analysis. From the QSAR equation it was found that
thermodynamic and electronic parameters show good correlation with biological activity. This
series of N-substituted 2, 4-thiazolidinediones may be potent alternatives to some of the related
antidiabetic agents
