The position of the front solvent is marked with a pencil and chromatogram and

then dried and sprayed with a ninhydrin solution. Ninhydrin reacts with amino acids to
produce colored compounds, mainly brownish orange or purple.

Ninhydrin test occurs when ninhydrin is heated with amino acids will form a
colored complex. Amino acids can be determined by kuntitatif by using the color intensity
formed is proportional to the amino acid concentration tersebut.Pada this reaction is
released CO2 and NH4 so that amino acids can be determined quantitatively by measuring
the amount of CO2 and NH3 released. Prolin and hydroxy proline produces complex colors
which is different in color from other amino acids. The color complex that forms contains
two molecules of ninhydrin which react with ammonia released on the amino acid
oxidation. The positive test results of the ninhydrin test are given on amino acids
containing -amino acids and peptides having - free amino.

Paper Chromatography on Amino Acids

Analysis of these amino acids is necessary for example to analyze industrial
outcomes such as food, fodder, medicines, as well as for the analysis of biological fluids
and protein hydrolysates (Rediatning and Kartini, 1987).
Amino acids that react with ninhydrin form a product called Ruhmann's purple.
This reaction is commonly used as a spotting test to detect the presence of amino acids on
chromatographic paper.
