By the end of this chapter you should be able to:
● use a reference source formula to calculate quantities of each ingredient
required to make a given amount of a pharmaceutical product
● correctly interpret formulae where ingredient quantities are listed as
parts or percentages.
Solution steps
1 As this is a liquid preparation, calculate how much of each
component is required to make 1 mL of the product.
2 Calculate, therefore, how much is required to make 300 mL of
the product.
Conc. 25 mL 25 ÷ 1000 mL (25 ÷ 1000) 7.5 mL
Chloroform × 300 mL

Example 4.2 You are required to prepare 5000 g Zinc Cream BP.
The formula is given below:
Zinc oxide 320 g
Oleic acid 5 mL
Wool fat 80 g
Solution steps
1 As this is a solid preparation, calculate how much of each
component is required to make 1 g of the product.
2 Calculate therefore, how much is required to make 5000 g of the
product.
Zinc 320 g 320 ÷ 1000 g (320 ÷ 1000) × 5000 g 1600 g
oxide
In the above two examples, the final line of the reference formula indi-
cates that the product is to be made up to a certain weight or volume.
However, this may not always be the case. Consider the formula for
Hydrous Ointment BP in Example 4.3. The total weight of the ingre-
dients is 1000 g, so the quantities of each ingredient must be scaled up
or down, based upon how much of the product is required.
Phenoxyethanol 10 g
Solution steps
1 Add together the quantities of each ingredient.
2 Divide the quantity of each ingredient by this sum to find how
much of each ingredient would be required to make 1 g of the
product.
3 Calculate the amounts required for 50 g.
Total weight of the ingredients is 1000 g. Therefore:
Wool alcohols 500 g 500 ÷ 1000 g (500 ÷ 1000) × 25 g
ointment 50 g
Solution steps
1 Work out the quantities of each ingredient that would be
required to make 100 g of the product – these will be the same as
the percentages.
2 Calculate the amounts required for 30 g.
Hydrocortisone ointment 25% 25 g (25 ÷ 100) × 30 g 7.5 g
Solution steps
1 Sum the total number of parts.
2 Divide the number of parts of each ingredient by this sum to
obtain a fraction.
3 Multiply this fraction by the required weight of the product to
find the weight of each ingredient required.
