ICSNR2017 Abstract
Ganoderma boninense is an economically importance pathogen in Malaysia due to the fact that the fungus
exerts one of the deadliest disease, Basal Stem Rot (BSR) in Malaysian oil palm plantations. There is an
urgent need to establish an optimum and suitable growth condition for G. boninense in laboratory
especially when it comes to the development for control strategies against the pathogen. In this work, the
interaction between types of media (Potato Dextrose Agar (PDA), Malt Extract Agar (MEA), and Rose
Bengal Agar (RBA)), temperature of incubation, and pH of media was investigated to optimize critical
conditions for mycelium biomass production and growth rate of G. boninense using Response Surface
Methodology (RSM). A central composite design (CCD) was applied and quadratic polynomial regression
model was used to analyze the experimental data using analysis of variance (ANOVA). ANOVA analysis
showed that the model was very significant (P<0.0001) for both mycelium biomass production and growth
rate of the fungus. The quadratic polynomial regression model suggested that the maximum mycelium
biomass yield of 2.91g, and the fungal maximum growth rate at 11.90mm/day can be obtained when G.
boninense was cultured on PDA, and the optimum temperature and pH were set at 27.9OC and 5.6
respectively. In conclusion, this work found CCD and RSM were very effective in optimizing G. boninense
culture conditions. G. boninense biomass production and growth rate were greatly influenced by the types
of media used, temperature of incubation, and pH of media.
