Inhibition of acetylcholinesterase (AChE), the enzyme in breaking down acetylcholine is

considered as one of the potential treatments in treating the symptoms of neurodegenerative

diseases such as Alzheimer’s disease and other dementia. The present study was conducted to

evaluate the inhibitory activity of Arachis hypogaea Linn. and Canarium ovatum Engl. through

in-vitro using Acetylcholinesterase Activity Assay Kit (MAK119) from Sigma. Oil was

extracted from nut kernels and 8 concentrations (100 mg/ml, 50 mg/ml, 25 mg/ml, 12.5 mg/ml,

6.25 mg/ml, 3.14 mg/ml, 1.56 mg/ml, and 0.78 mg/ml) were prepared. Hexane was used as the

blank and memantine as a positive control. The oil and AChE were added to the 96 multiplate

wells and the absorbance were read at 412nm using ELISA plate reader. Percent (%) inhibition

and half maximal inhibitory concentration (IC50) were obtained. Results obtained from the study

showed that both oils have inhibitory activities against AChE having percent inhibition from (put

ranges po do not specify conc). Both of the kernel nut oils were significantly different (p>0.05)

with the blank control. In IC50 value, C. ovatum revealed the best inhibition potential with

01.878 μg/mL in comparison to A. hypogaea with IC50 of 2.821 μg/mL. In general, both oil

extracts used in this assay showed promising results especially the C. ovatum.
