New components, like ionic liquids (IL) and deep eutectic solvents (DES), have been recently
applied as phase forming compounds in aqueous two-phase systems (ATPS) to enhance the
capacity of these systems in the partitioning of a large range of biomolecules. The DES are
formed by an association of a hydrogen bond acceptor (HBA) and a hydrogen bond donator
(HBD), and are characterized by an easy preparation, high biodegradability and
biocompatility. Recently some studies have shown that there are no real DES-based ATPS,
due the break of the hydrogen bonds between the HBA and HBD in aqueous systems.
However, some authors showed that the presence of a fourth component can control the
properties of the coexisting phases and present a favourable influence in the liquid-liquid
equilibrium and biomolecules partition. Taking this into account, the main goal of this work is
the use of ethanol as a fourth component to evaluate its capacity of modulating the partition
behavior at the ATPS: cholinium chloride + ethanol + dibasic phosphate potassium + water.
Three molar ratios of ethanol to cholinium chloride were used (1:2; 1:1 and 2:1). The LLE
behavior (binodal and tie-lines) as well as partition coefficients of alkaloids, amino acids and
phenolic compounds were obtained. The ethanol influence in the partition of the biomolecules
is discussed and the capacity of the ethanol to modulate the partition was perceived.

Key-words: Deep eutectic solvents (DES), partition coefficients, biomolecules, cholinium

chloride.
