NOTICE
Analytical instruments for in vitro diagnostic application involve the handling of human samples and controls
which should be considered at least potentially infectious. Therefore every part and accessory of the respective
instrument which may have come into contact with such samples must equally be considered as potentially
infectious.
BIOHAZARD
The „BIOHAZARD“ warning label must be affixed to instrument prior to first use with biological material !
Servicing Note:
Before doing any servicing on the instrument it is very important to thoroughly disinfect all possibly contaminated
parts. Before the instrument is removed from the laboratory for disposal or servicing, it must be decontaminated.
Decontamination should be performed by authorised well-trained personnel only, observing all necessary safety
precautions. Instruments to be returned have to be accompanied by a decontamination certificate completed by
the responsible laboratory manager. If a decontamination certificate is not supplied, the returning laboratory will
be responsible for charges resulting from non-acceptance of the instrument by the servicing centre, or from
authority’s interventions.

1 Introduction
The HumaLyzer 3500 is a semi-automated, programmable photometer intended for In Vitro Diagnostic (IVD) use by
qualified laboratory staff only.
For best results become familiar with the instrument and its capabilities before attempting any clinical diagnostic
tests. Refer any questions to your HUMAN distributor.

WASTE OUTLET: Before switching on the instrument, remember to connect the plastic outlet connector (red) to a
waste tank by means of a silicone tube.

2.3 Location of the Instrument
The instrument should be located in a clean environment, placed on a stable surface, and away from direct
sunlight, which could affect the operating temperature and the quantity of light measured by the instrument.
2.5 Caution
Do not connect the instrument to a power supply different from the value indicated on the label.
- Before connecting the power and finishing the installation section, make sure that the instrument is turned
off (check the polysnap located on the rear part of the instrument).
- Make sure that your AC main line has an efficient ground line. A bad ground line connection may
compromise analysis results and damage the instrument.
- After turning on the instrument, pay attention not to spill liquids or micro solid substances on the surface
around the instrument.
- Keep the instrument away from young children.
If the above procedures are carefully followed, it will be possible to TURN ON the instrument by using the switch
located on the polysnap block.
- Peristaltic pump that allows the solution to enter the instrument for measurement.
- Incubator well which keeps the flow-cell or manual cuvette at the temperature of approximately 37° C.
- Inlet tube.
Make sure that the transparent tube (the one that connects the flow-cell with the internal hydraulic circuit) is not
twisted or squashed.
Place the red tube around the peristaltic pump and make sure that the white plastic connections are set as shown
in the figure below:

Under the measurement block there is a lever (the PUSH button). Press the lever for no more than half a second to
pump samples into the flow cell.

2.8 Incubator
The incubator temperature is brought up to 37°C by the software and it will remain constant until the instrument
is turned off.
NOTE: It is very important to heat the macro cuvettes to the proper temperature for obtaining the most accurate
analysis results.

(*): This menus are dedicated for internal company testing procedure. No intended use for service.
1. Press the “SEL. FILTER” message and the sub-menu will appear.
2. Use the right and left arrows to select a specific filter wavelength, and then press Exec.
3. The filter wheel will rotate in order to set the selected filter under the light path.

4.2 T. CUVETTE
Press the “T. CUVETTE” message and the sub-menu will appear.
The 1° row stated as: “Temp. Cuvet” will show the flowcell current temperature.
In case the temperature need adjustment, proceed as follow:
This screen allows you monitoring temperature gradients of incubator and cuvette according to temperature
displayed in user program and digital level acquired by microcontroller (both current and target).
4.3 T.INCUBATOR
Press the “T.INCUBATOR” message and the sub-menu will appear.
The 1° row stated as: “Temp. Incub.” will show the Incubator block current temperature.
In case the temperature need adjustment proceed as follow:
- Select Filter: The filter that operator want to set, use the right and left arrows to select: (340nm, 405nm,
505nm, 546nm, 578nm and 630nm)
- Actual Filter: The filter currently placed under the light beam
- Absorbance: The Abs value of the substance present in the flow cell.
- Tension: The mVolt value present on the Photodiode preamp.

4.5 Lamp Targeting
DO NOT TOUCH THE LAMP DIRECTLY WITH THE HANDS!
1. Select filter 340, then press the wheel button and make aspiration with distilled water
2. Set the test tube of distilled water in the inlet pipe and press the PUSH button.
3. After a few seconds, continuous evolution of the tension value will appear on the display.
4. Loosen the screws of the lamp stirrup
5. Target the light bundle trough the reading hole to obtain the max tension on the display
6. Screw the screws
1. Select filter with arrows then press wheel icon, the desired filter will be placed under the light beam
2. In order to obtain the Abs value make and aspiration with water and wait for measurement.
3. Set the test tube of water (or other substances) in the inlet pipe and press the PUSH button.
4. After a few seconds, continuous evolution of the ABS value will appear on the display.
5. Press 0 key to have zero on the current ABS value (water or other substances). You will choose this
option when you will set to zero the instrument.
6. Prepare a new sample (reagents, potassium-dichromate or others) and press the PUSH button to start a
new ABS measurement session.
7. At the screen exit, the instrument expels the volume from flow cell via the outlet waste.
Tension Mode Measurement is very useful to verify lamp calibration, filter / lamp / lens / photodiode / preamp
board quality, position sensor working.
Moreover in this screen you can test correct pump motion.
If you have a correct calibrated optical group, when you use the flow cell, the TENSION values are between 250
mV (for 340 nm) and about 3.5 V (for 630 nm) for all filters.
4.6 WASHING
When pressing this message the peristaltic pump will be activated for around 10 seconds.

Pump Calibration
- Exit from service and from main menu enter in setup menu
- Look for “Automatic empty” menu and insert 5000 value (air gap delay time, equal to 5 seconds), then
save. See also user manual.
- Enter in Run method and run glucose.
- Prepare 500 l of water in a cuvette set the aspiration tip in a bottom angle of manual cuvette and press
aspiration button.
- If the pump works correctly, the water should be aspired completely from the manual cuvette, and
water should stop at the top of the aspiration tip.
- After 5 seconds the air gap will be aspired and liquid front will rise up.
- Repeat operation several times

4.9.1 Humalyzer Hydraulic Layout:

After the complete aspiration (sample aspiration + air gap), if water stops after at beginning of aspiration tip or
stops to near the cuvette the red tube has a problem and you have to adjust aspiration by changing the aspiration
pump steps.
Scroll down to the Touch screen Calibration menu and select it:
Inside menu Touch with a pencil and an indicator will appear in correspondence of the pencil tip.
The tip of the pencil must be in the same position of the indicator, try different position of touch, especially the one
where you report the problem on keyboard.
In case the tip and indicator are too far, you need to recalibrate touch screen.
Press Door to exit

4.10 HOST
A Host (program manage Running under Windows XP and supplied separately) to operate the Humalyzer from an
external PC.
In the Host program it is located all the features of the HumaLyzer software, plus an extended database and a large
choice of printer will able to be used with Humalyzer.
In order to connect the Humalyzer to a Host Computer via RS232 proceed as follow:
Check Voltage between the yellow wire and the black wire of J26 to measure about 13.5 Volt

- Open case of the instrument, pulling out four screws down the equipment and six ones on the
back side.
- Unscrew the four screws around the cuvette site, and move away pump support (the one with
pump rotor).
- Remove the lamp protection box by unscrewing the two screws.
- Move away the little tongue that fixes the lamp, by pulling out the only screw.
- Extract old lamp, and set carefully the new one, bringing it up to the bottom of the site (pay attention to
not touch lamp glass with finger skin).
- Set up again the little tongue, and check the top of the lamp being inside the little hole.
- Perform Lamp Targeting (see pag.14)
- Set up again the lamp protection box
- Set up again pump support and equipment case.

8 MAIN BOARD
- The Touch Screen board carries a “Managing Firmware” of the instrument. This firmware can be loaded
directly from serial port present in the rear panel. The 99% of upgrades will be performed on this board.
- The main board controls all the peripherals (device), frequency of re-programming for the “Peripherals
Firmware” is rare. In order to upgrade the it is necessary to open the instrument.
In order to upgrade the instrument firmware, please follow step by step the bellow stated instruction:
1. Connect one side of the re-program cable to one of the available PC COM sockets and the other side to
the HumaLyzer RS232 sockets
2. Close the contact present on the re-program cable PC/instrument.
3. Switch on the instrument.
4. Start on PC the program FlashSta and perform the following operations:
Select the COM to which the re-program cable has been connected, then press OK

Select the file to load (demo.mot)

Now the programming mode start, it will last some seconds and on the PC screen will appear the following:
The programming time is displayed by the blue bar; at the end of programming the blue bar will be complete.
NOTE: the exit from the program is not immediate, a delay of few seconds will be needed.

In order to upgrade the instrument Assay parameters and make a backup of all the assay parameters present inside
the HM memory, please follow step by step the bellow stated instruction.
In order to operate these procedure the relevant program named SetupSmpDownload.exe must be installed in the
PC (the PC must be RS232 connection equipped)
- In order to install SetupSmpDownload.exe, copy it on desktop then bobble click and follow installation
indications
- After the installation is complete an icon will appear on desktop
- Click on icon and the following program will appear:
- Select Com by pressing Com button in case the PC has more than 1 com connection
- Select File, open the file and select the .Met(this is Human GmbH assay parameters adaptation) or other.
- In case no file is present in Select file option, go in C\HM\Method and copy the file

8.2.1 Download Procedure
This program allows to load in the HM automatically methods parameters from a PC via RS232.
1) Connect one side of the re-program cable to one of the available PC COM sockets and the other side to
the HM RS232 sockets
2) Open the contact present on the re-program cable PC/instrument.
3) Switch on the instrument.
4) Start on PC the program SmpDownload.exe
5) Select File, open the file and select the.Met file, than press OK button
6) In the HM enter in service menu and select Download
7) In the SmpDownload program press Download button
8) The methods will be loaded into the Hm automatically
The same program allows to record the methods parameter stored inside a HM in the PC via RS232.
1) Connect one side of the re-program cable to one of the available PC COM sockets and the other side to
the HM RS232 sockets
2) Open the contact present on the re-program cable PC/instrument.
3) Switch on the instrument.
4) Start on PC the program SmpDownload.exe
5) Press File name button and name a file where all the assays parameters present on HM will be stored in
PC, then press Ok
6) In the HM enter in service menu and select Upload
7) In the SmpDownload program press Upload button
8) The methods of HM will be loaded into the PC automatically and stored in C\HM\Method

12. with the tip of pencil touch the middle of the flashing indicator until it disappear
13. Repeat this operation for all the indicator that will appear in sequence (16)
14. Exit and the tip of the pencil must be in the same position of the indicator, try different position of
touch, especially the one where you report the problem on keyboard.
15. If touch screen is now calibrated press SAVE button or F10 (with external keyboard)
16. If still touch screen is still needing adjustment repeat operation from point 1
Inside menu Touch with a pencil and an indicator will appear in correspondence of the pencil tip.
