ABSTRAC

The analgesic and nonsteroidal antiinflammatory drugs share certain therapeutic


activities and side effects, their mechanism of action is mediated by the inhibition of
cyclooxygenase (COX 1 and 2), the enzyme responsible for the biosynthesis of
prostaglandins and related autacoids. In Unit coordinator Pharmacovigilance (UCNFv) a
periodic analysis of suspected adverse reactions reported is made and in 2001, it was
observed that of the 10 drugs suspected of producing greater number of adverse
reactions 50% corresponds to the group of analgesics and anti-inflammatory drugs
(NSAIDs) which is in turn one of the largest consumer groups in the country. Among
the elements most commonly used drug in the world to control mild to moderate pain
include nonsteroidal antiinflammatory drugs (NSAIDs). Recent data on the
cardiovascular safety of these drugs, combined with gastrointestinal and renal risks
already known, require updating and reconciling the vast amount of information that
annually receive about these compounds. It is also imperative that work on research
carried out within the framework of valid scientific evidence and better designed
studies, properly selecting the comparators drugs, without bias, avoiding as far as
possible conflict of interest. Preferred selective NSAIDs and cyclooxygenase-2
(COX-2 or coxibs) have failed to meet optimistic expectations awakened by its entry
into the global pharmaceutical market, so we have to wait for the "superaspirin". The
objectives of this paper are to review available information on the efficacy,
effectiveness and safety of these therapeutic compounds .; improve our understanding
of their mechanisms of action and publicize the latest provisions and
recommendations of the competent authorities, for a more rational use of the same
prescription.
