Dissection of brain and measurement of morphological parameters:
After proper washing with normal water, weight of the whole brain (fresh) will be taken with the help of weighing machine. Whole brain will be then freely suspended in 40% formaldehyde solution for two weeks for fixation and subsequent study. Fixed sample will be initially washed with the free flowing tape water to wash away the formaldehyde solution to avoid irritation of the eyes and nasal mucosa by the formalin vapor. Fixed brains will be inspected visually for gross morphological abnormalities. For the preparation of the specimen, the sample will be taken in a metallic tray and the brain stem will be sectioned at the level of the cerebral peduncles and the hemispheres will be separated by a midline sagittal section with a B.P.blade and then following measurements will be taken - - Weight of the cerebral hemisphere will be measured with the help of a weighing machine.(left and right cerebral hemispheres were weighted to the nearest 10 mg on a Sartorius 2842 balance Henery 1989 lit-2 ) - Length of each cerebral hemisphere will be measured with the help of a metallic scale graduated in cm. The length will be measured from frontal pole to occipital pole. (distance between coronal tangents to frontal and occipital poles. Henery 1989 lit-2) - Maximum width (greatest distance between sagital tangents to medial aspect of hemisphere and temporal lobe Henery 1989 lit-2) and height (greatest distance between tangents to lowest point on the temporal lobe and highest point on medial border . Henery 1989 lit-2) of cerebral hemisphere will be measured with the help of a slide calipers. Then the piamater and the blood vessels will be carefully removed with scissors and dissecting forceps to expose sulci allowing better visualization. s
- Distances of the end of central, precentral, postcentral, lateral sulci from
frontal and occipital poles, distance between temporal pole to occipital pole, distances from end of central and lateral sulci to bottom of temporal lobe will be measured with the help of a slide calipers. - Then the length of central sulci, length of lateral sulci will be measured with the help of a slide calipers. -Lateral sulci –SF1: from the rostral end of the anterior horizontal ramus (a)to the end of the posterior horizontal ramus(b) SF2: from the rostral end of the anterior horizontal ramus to the end of the posterior ascending ramus
- Depth of the central sulci will be obtained by measuring distance
between external and internal edges of the sulcus. It will be measured with the help of wooden stick and measuring tape. - Thickness of central sulcus will be obtained by measuring the cortical thickness along the anterior bank of the sulcus and along the posterior bank of sulcus and cortical thickness will be estimated as the gray / white boundary and the outer cortical surface. - Angle between superomedial margin and central sulcus and angle between posterior border of lateral sulcus and inferior border of temporal lobe will be measured with the help of Goniometer.
