Experiment No. 1
Benedict’s test for Simple Reducing Sugars

Abstract

This activity will help determine the amount of sugars in a test sample. In 8 test tubes, 1 ml
of each test samples were added to 2 ml of Benedict’s solution. The 8 test tubes were heated for
2-3 minutes afterwards. The experiment revealed that among the 8 test samples the orange juice
and colorless soda contains a lot of sugar, while the milktea and beer contains little sugar.

I. Introduction
This experiment will show the presence of reducing sugars in a test sample. A color change
from blue green to brick red will indicate an amount of reducing sugar in a test sample.

II. Methodology
2 ml of Benedict’s solution was added in 8 clean test tubes. After this, 1 ml of each test
sample was added to the test tube containing the Benedict’s solution. The test tubes containing
the test sample and solution were heated for 2 to 3 minutes. Color changes were noted according
to the table below.

Test 1.

Based on Figure 2, water, corn starch solution, milk, and cooking oil did not show a color
change. On the other hand, orange juice, colorless soda, milktea and beer had a color change.

IV. Conclusions and Recommendations

Based on Figures 1 and 2, it can be concluded that the orange juice, colorless soda, milktea
and beer contains simple reducing sugar where orange juice and colorless soda contains the
highest amount of sugar meanwhile the milktea and beer had a little amount of sugar.
The consumption of drinks like orange juice, soda, milktea and beer may increase the
amount of sugar in person’s body. In recommendation, moderating the consumption of the drinks
mention may help in reducing the amount of sugar intake.

Experiment No. 3
Iodine Test for Starch

Abstract

This experiment will show whether starch is present in a test sample. In 8 test tubes 1 ml of
each test samples were added to 10 drops of Iodine solution. After this, the test tubes were
heated for 2-3 minutes. If a test sample contains starch, it can be revealed because it will exhibit
a violet-black color change because the iodine solution was added. This experiment revealed
that cornstarch solution contains starch.

I. Introduction
Iodine Test for Starch will determine whether starch is present in a particular test sample. If
a violet-black color change is observed, it indicates a positive result.

II. Methodology
10 drops of Iodine solution was added in 8 clean test tubes.1 ml of each sample was
added to each test sample with occasional shaking. Violet to black color change were noted.

III. Results and Discussion

Out of the 8 test samples, 7 did not have any color change which indicates a negative result.
Only the cornstarch solution exhibit a color change which indicates positive results.

IV. Conclusions and Recommendations

As shown in Figure 1, it can be concluded that water, orange juice, colorless soda, milk
tea ,milk, beer, and cooking oil does not contain starch and only the cornstarch solution indicated
its existence.
Average amount of usage of corn starch will lessen the buildup of fats since corn starch has
a sugar, in recommendation to prevent increasing of sugar in our body we need to practice proper
usage of it.

Experiment No. 4
Biuret’s Test for Proteins

Abstract

This activity will help us to determine if there is a presence of polypeptide and protein. In
order to conduct this activity 8 test tubes each containing 1 ml of 8 test samples will be prepared.
After that 10 drops or Biuret’s solution will be added. If there will be a change of color to purple
or pink it means that there is a presence of polypeptide. If the color changes into violet it only
indicates that the test sample has a presence of protein. The experiment revealed that the fresh
milk contains polypeptide.

I. Introduction
Biuret test is done to determine the presence of protein and polypeptide. If the color is
changed into pink or purple, it indicates that the test sample has polypeptide. On the other hand,
if the color is changed in to violet then it indicates that the test sample has whole protein.

II. Methodology
8 test tubes were labeled according to each sample, a total of 1 ml of each test sample is
added to the test tubes according to label then 10 drops of Biuret’s solution is then added. After
that, the solution will undergo the process of shaking. The color changes is recorded after the
experiment is done.

III. Results and Discussion

Out of all the 8 test tubes, only fresh milk had shown change in color that turns in to purple
which indicates a positive result that milk has polypeptide, while the rest test samples had
negative.

IV. Conclusions and Recommendations

According to the results that we have gathered, we can conclude that the fresh milk has a
presence of polypeptide based on the change of color. On the other hand, the other samples
doesn’t show any presence of polypeptide because they didn’t show any signs of change in color.

In recommendation, if you want to assess the concentration of proteins. It is advisable to do

the Biuret test. Because it is easy to do and easy to determine only by determining the change
in color.

Experiment No. 5
Grease Spot Test for Lipids

Abstract

This activity will help determine if a test sample is lipid or not. By putting 1 drop of each
sample on the surface of an unglazed paper, we were able to determine that the cooking oil is
lipid because it left a translucent mark.

I. Introduction
Lipids makes an unglazed paper transparent in front of a light source. Through the Grease
spot test it can be determine if a test sample is a lipid or not. A substance must be able to leave
a translucent (semitransparent) on the unglazed paper in front of a light source for it to be
consider as lipid.

II. Methodology
A drop of each sample was put on the unglazed paper and was drawn a circle on each spots
afterwards. After it dried, the unglazed paper was held in front of a source of light and the results
were recorded.

III. Results and Discussion

IV. Conclusions and Recommendations

Too much lipid in the body can cause different diseases to our heart. Based on the results,
cooking oil was concluded as a lipid. Therefore, cooking oil should be not be consume
occasionally. It must be used moderately to prevent our body from developing further
complications to our body.
