LABORATORY EXERCISE 2

THE PLANT CELL

TABLE OF CONTENTS
INTRODUCTION

According to Soft Schools 2019, plant cells are type of eukaryotic cell that are

found in the organisms within the Plant Kingdom. Plant cells are differ from other cells of

organisms because of the presence of chlorophyll. It is the green substance or organelle that

makes food through the process of photosynthesis. Study shown that plant cells also contains

large central vacuole that serves as a holding space for other materials and organelles in the plant

cell.

In observing a cell, it is a must to use a microscope . A microscope may be used to

look at the anatomy of small organisms such as insects, the fine structure of rocks and crystals,

or individual cells (Sciencing, 2019).  Microscope has different magnifying lenses that helps to

view the different parts of the cell. It is needed to have a proper knowledge before using. Also,

knowing the different parts and function of microscope is needed especially the different

magnifying lenses and the different amount of magnification in its lens.

The visibility and movement of the chloroplast in the cell is possible under the

microscope with the use of different magnifying lenses. Plant chloroplasts are commonly found

in guard cells located in plant leaves. It moves around in the process of photosynthesis.

In this laboratory work, the students saw the difference and similarities of the different plant

specimen, the movement of chloroplastid is seen and the reaction and the structure of starch

under the microscope.

Materials
The researchers used Compound Microscope, Glass Slides, Cover Glasses, Tap Water, Weak

Iodine Solution, and Razor Blade. With a specific specimen of Bell Pepper, Hydrilla, Onion, and

Potato. Most of the materials specifically the equipments used are from the school supply room

and the specimens are provided by the researchers.

Methods
The researchers used different methods on each specimen.
A. Onion
First, researchers carefully peeled off a portion of the outermost layer of the epidermis of the
onion bulb. The specimen was placed on the cover slide. Amount of water was dropped before
the cover slip was placed. The researchers examined the specimen under LPO and HPO lenses.
In another slide, a weak iodine solution was dropped into the specimen. Lastly the specimen with
iodine solution was examined under LPO and HPO lenses.
B. Hydrilla
First, the hydrilla plant was placed under the presence of light for 30 minutes. After that, a leaf
was taken and placed in a slide. A right amount of water was dropped in the specimen before
putting the cover slip. The arrangement of cells was observed under the LPO lens of the
microscope. Also, under the HPO lens, the researchs are able to identify the different parts of a
single hydrilla cell. Lastly, the movement of chloroplastid was seen under the microscope.
C. Potato
The researchers cut a right amount of potato tuber and placed in two different slides. The cover
slip was placed after putting a drop of water. The specimen was observed under the LPO and
HPO lenses. On the second slide instead of water, weak iodine solution was applied. Like the
first potato specimen, the slide was observed under HPO and LPO lenses.
D. Bell Pepper
Using a razor blade, the specimen were cutted and placed in 2 different slides. Water was
dropped in the first specimen and iodine solution on the second. Both are observed under the
HPO and LPO lenses of the microscope. The chromoplastids were observed within the cells. The
images seen under the microscope were drawn and labeled.
