GENERAL SYSTEM INTRODUCTION
1-1 SYSTEM DESCRIPTION
‘The monochromator on the Model 340 Spectrophotometer consists of a plane diffraction grating
in an {7 Ebert mounting with first surface reflective optics. This design ensures efficiency
and gives the 340 a continuous wavelength range of 330nm to 1000nm to cover most routine
ULV. and colorimetric tests. This range can be expanded to 195nm with a Far U.V. accessory.
Only three operator controls make the 340 one of the simplest spectrophotometers in the
world to use. A large 3 1/2 digit LED display provides easy readability to minimize errors.
With a simple turn of a knob, the readout can be switched from percent transmittance to
absorbance. It's so easy, the Model 340 requires virtually no training time.

2-2 Instrument Checkout
After the 340 is unpacked inspect it for physical damage. Report any observed damage
immediately to the Freight Carrier and your dealer.
2-3 Installation
‘A. The 340 requires either 100, 120, 220 or 240 VAC at 50 Hz or 60 Hz from a properly
wired and grounded, single phase power outlet. The 340 is delivered preset to 120 VAC.
Verify which voltage is present and set the voltage selection board on the back panel (see
FIG 2-1) to the appropriate setting as follows:
1. Locate power inlet connector on back panel.
2, Be sure the power cord is not installed and slide the plastic protective cover to
the left exposing the voltage selector board. If the voltage shown is incorrect, pull
back the black fuse lever to remove the fuse and remove the board by pulling it
firmly with a set of small pliers.
3, Turn the board until the proper voltage is facing you and reinsert the board. The
proper voltage is now visible from the rear of the instrument.
Insert the proper fuse for the voltage selected.
For 100 and 120 VAC use 0.6 Amp Slow Blow fuse. (installed)
For 220 and 240 VAC use 0.3 Amp Slow Blow fuse. (included in
accessories)

CAUTION
‘Two spare fuses are provided with the instrument, 0.3 Amp and 0.6
Amp. In addition, the instrument comes with a 0.6 Amp fuse
installed for 100 and 120 VAC operation, In areas where 220 and
240 VAC are used, the installed fuse and the spare 0.6 Amp fuse
should be discarded and replaced with the 0.3 Amp fuse.
5. Slide the cover to the right.
B, Locate the instrument on a vibration-free laboratory surface. Allow two inches behind
panel for air circulaton and power cord.
C. Connect the power cord to instrument and to the proper outlet after verifying that the
voltage of the instrument has been properly set.
D, Turn power switch (left hand knob on front panel) to Transmission Mode. The instrument
panel should illuminate, If the display does not illuminate, immediately turn the instrument
off and consult the troubleshooting section (see section 5-2).
E. Adjust lamp for maximum output (see section 4-4).
F, Refer to section 3 for operational details of the 340.

OPERATION
3-1 Deseription
Refer to FIG 3-1 for location of the components.
A. Controls and Indicators:
1. MODE SWITCH - OFF - Turns instrument off.
TRANS - Turns power on and places instrument in transmission mode.
ABS - Turns power on and places instrument in absorbance mode.
2. ZERO - When instrument is in TRANS mode and the ZERO SET button is pressed and
held, this’ knob is used to set the display to 0.0 % T.
3. 100%T/0A - In TRANS mode is used to set 100% T on the display.
In ABS mode is used to set .000 A on display.
Consists of two knobs, a larger knob next to the panel for coarse setting and a smaller
knob in front for fine setting.
4, DISPLAY - 3 1/2 digit display to indicate transmission or absorbance.
5. TRANS - Indicator LED lights when the instrument is in transmission mode.
6. ABS -Indicator LED lights when the instrument is in absorbance mode.
7, WAVELENGTH READOUT - 3 digit readout of the wavelength in nanometers,
8. WAVELENGTH ADJUST - Thumbwheel knob to set the wavelength of the instrument.
9, ZERO SET - Button is pressed when the instrument is adjusted to 0.0% T with the
ZERO knob. -
B, Cuvette Holder:
The standard cuvette holder supplied with the instrument takes a 13mm by 100mm tube,
To insert tube, lift cover from the front and insert tube until it reaches the bottom.
Close cover. See your dealer for optional cuvette holders available for your 340.
C. Stray Light Filters:
For optimum performance, a set of stray light filters are supplied, covering the wavelength
range of 330 to 1000nm. To change filters, push back handle on filter slightly to disengage
lock and lift straight up. Insert proper filter (it will automatically lock into place). The
instrument must have a stray light filter in position for proper operation.

3-2 Operation
Perform proper installation steps (see section 2) before instrument is operated for the first time.
A. Turn mode switch to TRANS, Allow a 15 minute warm-up.
B. Adjust instrument to desired wavelength.
C. Insert appropriate stray light filter into position.
D. Place cuvette with water or other blank solution into cuvette holder.
E. Press and hold ZERO SET button while adjusting ZERO knob until display indicates 0.0.

1, For transmission measurements:
a. Set MODE switch to TRANS.
b. Adjust 100%T/0A COARSE knob to approximately 100.
¢. Adjust 100%T/0A FINE knob to exactly 100.0.
d. Replace cuvette with sample cuvette and read transmission from digital display.
2. For absorbance measurements:
a, Set MODE switch to ABS.
b. Adjust 100%T/0A COARSE knob to approximately .000.
¢. Adjust 100%T/0A FINE knob to exactly .000.
d. Replace cuvette with sample cuvette and read absorbance from digital display.
33 Analog Output
External devices, such as data processing instruments or strip chart recorders, may be interfaced
to the 340, Spectrophotometric analog output is on the rear panel (see FIG 2-1). The zero to
one volt DC output is equivalent to the transmittance of the sample whether the 340 is in ABS
or TRANS mode.
Connection is made to the 340 with standard banana plugs. Observe the polarity as marked
on the panel.

PREVENTIVE MAINTENANCE
4-1 General
‘The 340 has been designed with the same reliability inherent in previous TURNER spectro-
photometers but with modern solid state electronics. It requires minimal preventive mainten-
‘ance and only the care one would give any quality instrument. It should be kept clean and out
of dusty areas or those with temperature and humidity extremes,
4-2 Fuse Replacement :
‘The fuse is mounted in a holder which prevents access unless the power cable is removed.
First turn off instrument and then remove cable from power outlet.
‘A. Remove power cable from back of instrument.
B, Slide plastic cover to left exposing fuse holder.
C. Locate black lever marked FUSE PULL. Swing lever out and to the left.
D. The fuse can now be removed.
E. Replace the fuse only with the proper Slow-Blow type. If voltage is changed refer to
installation instructions (section 2-3) .
0.6 Amp for 100 and 120 VAC applications
0.3 Amp for 220 and 240 VAC applications
F, Insert fuse and push lever back into place.
G. Slide cover to right and replace power cord in instrument and then into power outlet.
4-3 Cuvette Holder Removal
SEQUOIA-TURNER manufactures a variety of cuvette holders to increase the versatility
of the 340. The holders are easily removed as follows:
A, Lift the cover on the cuvette holder. Locate the Allen screw at the inside back of
the cuvette holder and about 1 em (.4") down.
B, Insert the 1.6mm (1/16") Allen Wrench into the screw and loosen it several turns (CCW).
C. Lift the cuvette holder out of the instrument.

D, Reverse the steps above to install the alternate cuvette holder.
4-4 Lamp Replacement
A. Turn the instrument off.
B. Locate the two thumb screws on the lamp housing on the right side of the instrument.
C, Remove the two thumb screws and remove the lamp housing cover.
D. The lamp is removed by pressing it toward the rear of the instrument while turning it
counter-clockwise.
Install a new lamp (Sequoia-Turner part number 330-058). Clean the lamp with lens
tissue.
F, Align the lamp.
1, Turn the instrument on,
2, Set the instrument in TRANS mode and adjust the wavelength to 340nm with the
UV (330-385nm) stray light filter in place.
3. Adjust the 100%T/0A knob for about 50% Transmittance on the display. If it does
not affect the readout set it to maximum,
4. Grasp the black knob at the base of the lamp holder and move it until the display
increases. Continue adjusting until the display is at a maximum, The 100%T/0A
knob may have to be turned down to keep the display from overranging. The
filament should be kept in vertical alignment at all times.
8. When the reading is maximized, turn the instrument off and replace the lamp housing
cover.
4-5 Cover Removal
NOTE
This should only be performed under the direction of a qualified
service representative. There are no user adjustable or replaceable
parts inside this instrument.
‘A. Turn off instrument and remove the power cable.
B, Remove the cuvette holder (see section 4-3).
C, Turn the instrument over and remove the six screws holding the cover on.
D. Turn the instrument right side up and lift the cover off.
E, Reverse the above steps to replace cover.

‘TROUBLESHOOTING
5-1 General
‘The 340 has no user replaceable or adjustable components inside the cover. Do not remove
the cover unless specifically authorized by the dealer or the factory. If steps below do not
solve problem contact your dealer's service representative.
5-2 Troubleshooting
* Front panel does not illuminate -
1. Turn power off.
2, Remove power cord from instrument and main power line.
3. Remove and inspect fuse (see section 4-2). Replace if necessary.
4. Reinsert power cord firmly to ensure proper contact.
5. Turn instrument on and verify proper operation.
* Front panel illuminates, source lamp does not -
1. Turn coarse 100%T/0A knob fully clockwise.
2. If lamp still does not illuminate, replace and align lamp (see section 4-4).
3. Turn instrument on and verify proper operation.
* Front panel and source illuminate, display will not change -
1. Perform installation procedure (see section 2-3) and operation procedure (see section 3-2).
