Paper chromatography for the separation of components

Chromatography:

It is a technique for examination and separation of mixtures of chemical substances. It


facilitates purification, isolation, and comparison of various components. It can be
undertaken for all kind of volatile and soluble substances, organic and inorganic, polar
and nonpolar, in different quantities under various conditions. Different substances are
partitioned differently between two phases (one is stationary phase and the other is
mobile phase)

Paper chromatography:

A drop or spot of solution containing the mixture of substances to be separated and


applied near one end of filter paper piece. The paper is carried out into two ways

1. Ascending paper chromatography


2. Descending paper chromatography

Ascending paper chromatography:

In ascending paper chromatography the solvent is given in a pool at the bottom of the
vessel in which the paper is supported. The solvent rises up by the capillary action

Descending paper chromatography:

The solvent is in trough form in which the paper is placed. The solvent flows down the
paper by combination of gravity action and capillary action.

Requirements

Beaker, filter paper, capillary tube, inks

Procedure

Select the suitable size of filter paper and pencil.

Draw the parallel line to one end of paper.

A number of small spots are marked at suitable distance on this line at equal distance.
The sample no. or nature of sample to be applied is written against each cross with
pencil. A drop of each sample is spotted at appropriate portion with capillary spots are
allowed to dry until the solvent is completely evaporated. The end of paper near the
spot is immersed in a suitable solvent for certain period of time

The solvent is displaced from origin through capillary action.

Calculate the Rf value

Precaution:

Large spot should be avoided as they may lead to poorer separation.

The filter paper should be haug properly so that the separation is appropriate.

Do not immersed the spot in the solvent.
