Drawbacks of single compartmental model

• Single compartment model does not take into


account finer details of dynamics of solute
transport across various membranes
• Each of the compartments such as cell
volume, interstitial volume, etc can be further
compartmentalized

Generalized N compartmental system model

• Each compartment is characterized by a


conservation of mass differential equation
describing rate of change of solute.
• For N compartments there are N equations of
the general form:

An example two compartmental model
Consider a two compartment system for the
distribution of creatinine in the body. Creatinine
is a waste product of metabolism in the muscle
which is cleared from the body through urine.
Assume creatinine production in the muscle is qin
and is given by a step input. Assume that
creatinine is transferred from plasma to muscle at
a rate constant K1 and from muscle to plasma at
a rate constant K2 and removed from plasma
through urine at a rate constant K3. Estimate the
concentration of creatinine in the plasma
compartment as a function of time.

Non linearity in compartmental models
• In many systems the transfer rates are
dependent on solute in a compartment.
• Compartmental models can still be applied by
incorporating non linearity in the model.
