Experiment No. 5

CARBOHYDRATES

I. Objectives

1. To observe reaction of different carbohydrates using different test such as (a) Molish
Test (b) Benedict’s Test (c) Fehling’s Test (d) Barfoed’s Test (e) Picric Test (f) Iodine
Test (g) Seliwanoff’sTest (h) Phenylhydrazine
2. To examine the crystals formed under a low-power microscope
3. To draw and describe the crystals that is observe
4. To identify the unknown sugar

II. Procedures

I. Mollsch Test:
- Label six test tubes with number 1-6. Place 1ml of different sugar
solutions into five test tubes. Add distilled water to the sixth test tube
- xing.
- With the test tube held at a 45 degree angle, carefully and slowly run
10 drops of concentrated H2SO4 down the wall of the test tube so
that two layers form. Note the color at the point where the two layers
meet.

II. Benedict’s Test:


- Place 1ml of each sugars into seven tubes. Add 3ml Benedict’s
reagent into each test tube.
- Shake the solution and place all 7 test tubes in a boiling water bath at
the same time.
- Remove the test tubes after 5 minutes and allow them to cool. After
15 minutes, observe any changes that occurred.

III. Fehling’s Test:


- Dilute 2ml Fehling’s solution (a mixture of 1ml Fehling’s A and 1ml
Fehling’s B) with 8ml distulled water. Put 1ml of the diluted solution
into seven test tubes. Take 1 of the test tube, while heating on the
water bath, add xylose solution drop by drop and note the result.
- Repeat the test on the remaining test tubes.

IV. Barfoed’s Test:


- Prepare seven test tubes. Place 1ml of different sugar solutions. To
each solution, add 3ml of the Barfoed’s reagent.
- Shake each teast tubes. Place them in the water bath at the same
time. Remove them after 5 minutes, let cool.
- After 15 mins record your observation.

V. Picric Acid Test:


- Label seven test tubes. Place 1ml of the different sugar solutions.
- Add 1ml of picric acid solution to each test tube and add 5 drops of
NaHCO3 solution.
- Mix thoroughly then boil each test tube in the water bath for 1 minute
and observe the changes in color.

VI. Iodine Test:


- Prepare 5 test tubes. Place into test tube 1ml of the different sugar
solutions.
- To each test tube add 2 drops of Iodine solution. Note the color
formed.

VII. Seliwanoff’s Test:


- Prepare 5 test tubes. Place into each test tube 1ml of the different
sugar solution.
- To each test tube add 3ml of the Seliwanoff’s reagent and shake.
Place all test tubes in boiling water bath at the same time.
- Observe the changes in each test tube during the 15 minutes. Record
the color and time of its formation in each solution tested.
