TITLE Most Probable Number (MPN) for


Total coliform & E-coli

Introduction
The presence of coliform bacteria in the water indicates that the water may
have been contaminated with sewage and is not safe to drink. Regulations
require that coliform bacteria be completely absent from 95% of the water
samples.
Objectives

At the end of the laboratory activities, students would be able to:


1. To understand and implement suitable method to prove the existing
of E-coli and Total Coliform in water bodies.
2. To quantify the presence of E-Coli and Total Coliform in water
samples.

Learning Outcomes

At the end of the laboratory activities, students would be able to:


1. To identify and perform the appropriate experiment to
PREAMBLE determine the presence of total coliform & E-Coli in water
sample due to a given problem.
2. To analyze and interpret the data and identify the amount of
total coliform & E-Coli in different water samples.

Theoretical Background

The presence of microorganisms in water samples indicate that the


samples is harmful to be consumed. Microorganisms such as E-coli could
cause diarrhea and nausea. Measurement of E-coli and Total Coliform are
necessary to decide the appropriate amount of disinfectant.

PROBLEM Each group has to conduct an experiment to identify the amount of E-coli
STATEMENT and Total Coliform of a water sample.

Students are required to identify and summarize the methods/procedure


WAYS & MEANS
used to conduct the E-coli and Total Coliform and determine the presence
of total coliform & E-Coli in water sample. (Submit your list of apparatus &
sketched procedure before proceed with the experiment.

Each group is required to submit a technical report consists of introduction,
the apparatus used, the procedure undertaken for the test, data analysis,
RESULTS detail discussion on the obtained result with relevant standard and
conclusion.
Discussion on the delegation of work among the group members should be
recorded and attached in the report. (e.g. minute of meeting/screenshot of
your team discussion in WhatsApp group). References and standards
should also be attached together with the report.
The report must be submitted within 7 days after the completion of
the experiment.
