LABORATORY ACTIVITY 9
Test for Proteins

I. Desired learning outcomes


Proteins form the major portion of solutes in plasma. Serum proteins may be identified
by 2 general fractions-albumin and globulin. Albumin represents the most abundant protein
constituent, while the globulins are a heterogenous group comprising of immunoglobulins,
complement, enzymes hormones, coagulation factors and specific transport proteins.

After performing this activity, students should be able to:


1. differentiate the three types of proteins in terms of function, values and characteristics;
2. determine different pre-analytical, analytical, and post-analytical errors can affect protein
tests;
3. perform the procedure with care and accuracy; and
4. do mathematical computations to derive value of globulin and convert values obtain to
conventional units.

II. Materials
Photometer Albumin Reagent
Pipette Total Protein Reagent
Pipette Tips Syringe with Needle
Test Tube Cotton Balls
Small Beaker 70% Alcohol
Parafilm Folded Tissue Paper
Freshly Prepared 10% Bleach Solution
Serum/Plasma

III. Procedure
*This lab activity will be performed on 2 nd semester. Base your observation and discussion on
the video demonstration.

1. Serum or heparinized plasma may be used. Protein in the samples is stable for 8 days at
2-8˚C.
2. Bring sample and reagents to room temperature prior to use.
3. Label properly all test tubes.
4. Perform first the measurement for total protein:
a. Put 20 µL of sample and standard into each tube.
b. Add 1000 µL reagent to all test tubes.
c. Cover mouth of test tube with parafilm and invert to mix.
d. Incubate for 10 minutes at room temperature.
e. Read at 545 nm and record result. Take note that color is stable for 2 hours.

5. Perform albumin test:
a. Put 15 µL of sample and standard into each tube.
b. Add 1000 µL reagent to all test tubes.
c. Cover mouth of test tube with parafilm and invert to mix.
d. Incubate at room temperature for 1 minute.
e. Read at 630 nm and record result. Color is stable for 30 minutes.
6. Then determine the globulin level by getting the difference of total protein and albumin.
7. To convert the SI values back to conventional, divide the result by the following factors:
For albumin and globulin, divide the SI value by 10 to obtain the unit g/100mL

For total protein, divide the SI value by 10 to obtain the unit g/dL

8. Interpret the result using the reference values:


Total protein
Ambulatory: 64-83 g/L
Recumbent: 60-78 g/

Albumin
Newborn (2-4 days) 22-44 g/L
4 days to 14 days 38-54 g/L
Adult 35-50 g/L
>60 years 34-48 g/L

Globulin
1.1-2.5 g/L
