Activity 12
Qualitative Tests For Elements In Organic Substances

Directions: Using the internet, visit and educational website, and answer the
following questions.

Objectives:
At the end of this activity, the students should be able to:
1. Identify the different elements present in Organic substances.
2. Explain the results obtained in each test.
3. Appreciate how the tests are done.

1. TEST FOR THE PRESENCE OF CARBON AND HYDROGEN

a. Mix thoroughly 5 grams of CuO and 3 Grams of Sucrose in a dry ignition tube. Clamp
the ignition tube into the iron stand. Then, cover the ignition tube with a cork with an
attached bent glass tubing.

b. At the end of the glass tubing, place a beaker containing 50mL of a clear lime water or
Calcium Hydroxide.
c. Heat the mixture over a moderate flame. Observe and record the results.
Test For The Presence Of Carbon And Hydrogen
 Detected qualitatively by a heating a mixture (glucose) with dry CuO in a test tube.
 The glucose is oxidized forming CO2 and H2O.
 The presence of CO2 is indicated by the formation of a white ppt. of calcium
carbonate when gaseous products are passed through a solution of lime water.
 The presence of water is indicated by the formation of droplets of water in the cooler
end of the hard glass tube.

2. TEST FOR THE PRESENCE OF OXYGEN (FERROX PAPER TEST)


a. Prepare 4 pieces of dry 10mL test tubes and place the following reagents:

Tube #1 – 2mL Acetone Test Tube #3 – 2mL Benzene


Tube #2 – 2mL Isopropyl Acetone Test Tube #4 – 2mL Benzaldehyde

 What will happen to the compounds using Ferrox paper?

3. TEST FOR THE PRESENCE OF NITROGEN (SODALIME TEST)


Place 2 grams of Urea and a pinch of soda lime in a dry test tube. Heat it over a
moderate flame and note for the odor of the vapour produced. (Note: Do not inhale the
fumes directly)

Then immediately expose a piece of wet red litmus paper which is attached at one end of
the glass rod to the vapour produced. Note for the change of color the litmus papers.

4. TEST FOR THE PRESENCE OF HALOGENS


a. Beilstein Test

Heat the copper wire over a blue flame, continue heating until a luminous flame is
produced. Cool the wire and dip it in the beaker containing the chloroform solution.
Heat again over the flame. Repeat this procedure for 5 times. Observe and record
the results obtained.

b. Silver Nitrate Test

To 2mL of Chloroform, add 3 drops of concentrated Nitric Acid and boil gently for
few minutes. Add 6 drops of 2% Silver Nitrate solution. Shake the mixture. Note for
the formation of turbid or cloudy solution.

5. TEST FOR THE PRESENCE OF SULFUR (LEAD ACETATE TEST)

In a test tube, place 1mL of 5% albumin then, add 5 drops of 10% NaOH and 3 drops of
5% Lead Acetate. Place the test tube containing the mixture in a water bath. Then,
allow it to boil. Note for the color of the precipitate formed.
