CLASS:
Action An antibiotic that combines amoxicillin and clavulanic acid. It destroys bacteria
by disrupting their ability to form cell walls. - Clavulanic acid blocks the chemical
defence, known as beta-lactamase, that some bacteria have against penicillins.
Peak plasma time: 2 hr (amoxicillin); 1.1 hr (clavulanic acid)
Peak concentration: 8-22 mcg/mL (amoxicillin); 0.8-2.6 mcg/mL (clavulanic acid)
AUC: 40-80 mcg•hr/mL (amoxicillin); 2-6 mcg•hr/mL (clavulanic acid)
Half-life
Amoxicillin: 3.7 hr (full-term neonates); 1-2 hr (infants and children); 0.7-1.4 hr
(adults)
Clavulanic acid: 0.8-1.4 hr
Excretion: Urine, unchanged; 50-70% (amoxicillin), 25-40% (clavulanic acid)

Indication/ Indication
contraindication Infections due to β-lactamase-producing strains (where amoxicillin alone is not
appropriate) including resp tract, genitourinary & abdominal infections; cellulitis,
animal bites, severe dental infection w/ spreading cellulitis.

Contraindication
Hypersensitivity to penicillins. Cross-sensitivity w/ other β-lactam antibiotics eg,
cephalosporins. Previous history of jaundice/liver impairment associated w/
penicillins. Glandular fever or lymphatic lymphoma.

 
Side Effect/ Adverse Hypersensitivity reactions & GI disturbances. Reversible leukopenia &
reaction thrombocytopenia, thrombocytopenic purpura, agranulocytosis, anemia, slight
thrombocytosis, eosinophilia, abnormal platelet aggregation, prolonged bleeding
& prothrombin time. 

Nursing Consideration Report haematuria or oliguria as high doses can be nephrotoxic. - Assess
respiratory status. - Observe for anaphylaxis. - Ensure that the patient has
adequate fluid intake during any diarrhoea attack.
