Procedure for the lab

In this lab we will require that you write a procedure for the lab. The procedure that we want you to
write does not necessarily care about what you actually did in lab, like waiting around for other people
to finish playing around with the SEM, only what you should have done to take and image and examine
the factors that affect depth of field. To help you with this, this procedure has been written to outline
the steps of the lab that we want you to take away from it, and subsequently what we are looking for in
the procedure. It is still your responsibility to turn this bulleted list into eloquent, flowing prose, but at
least now you have all the steps we should have taken. Let me be clear: we are not looking for a
bulleted list in your procedure. You should not copy and paste this into your lab report as you will
receive no credit for that portion of the report.

 Preliminary preparation
o Mount sample on stage
o Pump down chamber
o Position sample in chamber
o Turn gun on
 Focusing sample
o Zoom in
o Adjust focus
o Repeat
o At ~10 um scale bar, adjust aperture
 Remove translation in wobble
o At ~300 nm, adjust stigmation
 Taking an image
o Increase scan speed
o Set averaging to line avg.
o Freeze image
o Save
 Tilt sample to 45°
 Change aperture to 120 μm, focus on center of image to get WD
o Focus sample
o Take image
 Change aperture to 60 μm
o Focus sample
o Take image
 Move sample down, refocus on center of image to get WD
o Focus sample
o Take image
 Change aperture to 120 μm
o Focus sample
o Take image
 Taking out sample
o Turn gun off
o Purge chamber
o Take out sample
o Pump down chamber
