EXPERIMENT 4- STOCK SOLUTION FOR MS MEDIA

Aim:
Preparation of stock solution for MS media (Murashige & Skoog media).

Principle:
PTC requires a continuous supply of certain inorganic chemicals besides carbon,
hydrogen & oxygen. The essential elements required in relatively large accounts are
turned as macronutrients which include N, P, K, Ca, Mg, S. in addition to
macronutrients plants also require traces of certain micronutrients like Fe, Mn, Xn,
Bo, Cu, Mo, Cl. Every time we prepare a media it is not possible to weight all salts
especially micronutrients since they are required in very small amount. Therefore
concentrated stock solutions can be stored for limited period in glass container &
used for reconstitution of media at the time of use.

Requirements:
Stock solution 1 (1x) – For macronutrients

CONSTITUENTS AMOUNT (in mg/L)

MgSO4.7H2O 370

K2H2PO4 170

CaCl2.H2O 440

Stock solution 2 (1x) – For micronutrients

CONSTITUENTS AMOUNT (in mg/L)

H3BO3 6.2

MnSO4.4H2O 22.3

ZnSO4.7H2O 8.6
Na2MoO4.2H2O 0.25

CuSO4.5H2O 0.025

CaCl2.5H2O 0.025

KI 0.83

Stock solution 3 (1x) – Important Ions

CONSTITUENTS AMOUNT (in mg/L)

FeSO4.7H2O 27.8

Na2 EDTA.2H2O 37.3

Stock solution 4 (1x) – Organic Nutrients

CONSTITUENTS AMOUNT (in mg/L)

Inositol 0.5

Thymine HCl 0.5

Pyridoxine HCl 0.5

Nicotinic acid 100

Glycine 2

Conclusion:
We have prepared stock solution of MS media (Murashige & Skoog media).
