Dose-Response relationships

It is the graphical representation of the dose of drug and its response is called dose response
relationships.
Types:
1. Graded dose response relationship .
2. Quantal dose response relationship

Graded dose-response relations:


As the concentration of a drug
increases the magnitude of its pharmacological effect
also increases. The relationship between dose &
response is continuous & gradual. A graph of this
relationship is known as a graded dose-response
curve. The curve can be described as a rectangular
hyperbola.

Importance:
This curve provides information regarding the potency, efficacy & selectivity of the drugs.

Quantal-dose response relationships:


This dose response relationship means the influence of the magnitude of the dose on the proportion of a
population that responds. For any individual the effect either occurs or it does not.
For example, a quantal dose response relationship can be determined in a population for the antihypertensive
drug atenolol. A positive response is defined as at least a 5 mm hg fall in diastolic blood pressure.

Importance:
1. It provide information regarding:
Potency of a drug
Efficacy of a drug
Selectivity of a drug
Variability of responsiveness among
individuals.
2. It also generates information
regarding the margin of safety of a
drug.
