SPECIFICATIONS
Method: Colorimetric (DR/900)
Range: 0.4 to 15.6 ppm Lignin
Testing Time: 40 minutes

DESCRIPTION
This test measures all hydroxylated aromatic
compounds, including tannin, lignin, phenol
and cresol. This method produces a blue color
proportional to the amount of these compounds
present in the sample. The results are reported as
total lignin expressed as ppm lignin.
The calibration used for Program Number 166
was prepared using Indulin® Kraft pine lignin
manufactured by Westvaco, Charleston Heights,
South Carolina.

INTERFERENCES
Sulfite interference is eliminated by adding 1 mL
of formaldehyde to the sample before testing the
sample.
Ferrous iron causes a positive interference. Two
ppm of ferrous iron produces a color equivalent to
about 0.5 ppm lignin. To eliminate interference of
ferrous iron up to 20 ppm, add one 0.2 gram scoop
of sodium pyrophosphate to the sample before
testing.

Important Note: See Procedure Step 4 for analyzing turbid samples.

Start Program Fill a clean sample cell (the Fill another clean sample cell Pipet 0.5 mL of TanniVer 3
blank) to the 25-mL mark (the prepared sample) to the Tannin-Lignin Reagent into
288 Lignin with demineralized water. 25-mL mark with sample. each cell. Swirl to mix.

Note: Filter samples

that are highly turbid.
Report test results as ppm
soluble lignin. Samples
containing large lignin
concentrations may
appear turbid, however
the turbidity could be
insoluble lignin. Mix the
sample well and make
a dilution to determine
total lignin. Do not filter.
(A few drops of 460-
S0279 may be added to
the sample to solubilize
the lignin. Sample pH
should be between 10
and 11 after addition of
460-S0279.) If soluble
lignin is to be determined,
filter the sample and run
this test on the filtered
sample. Again, make the
appropriate dilution.

Pipet 5.0 mL of Sodium
Carbonate Solution into each
cell. Swirl to mix.
Note: A blue color will
develop if lignin is present.

Remove the blank from
the cell holder. Place the
prepared sample cell into
the cell holder. Tightly cover
the sample cell with the
instrument cap.
Start TIMER 1
A 25-minute reaction period
will begin.
Read
Press: READ
The cursor will move to the
right, then the result in mg/L
lignin will be displayed.
Zero

Place the blank in the
cell holder. Tightly cover 0.0 mg/L LIG
the sample cell with the
