6 Novel Extraction Technique-Final
AIM: To extract the solute using microwave assisted extraction (MAE) and
ultrasound assisted extraction (UAE)
THEORY:
Microwave assisted extraction
Microwave as a source of heat
The mechanism of microwave heating is different from that of conventional heating.
In conventional heating, thermal energy is transferred from the source to the object through
conduction and convection. Here, microwaves penetrate into the pores and then
electromagnetic energy is transformed into heat through ionic conduction and dipole rotation.
Ionic conduction refers to the movement of ions in a solution under an electromagnetic field.
The friction between the solution and the ions generates heat. Dipole rotation is the
reorientation of dipoles under microwave radiation. A polarized molecule rotates to align
itself with the electromagnetic field at a rate of 4.9 X 109 times per second. The larger the
dipole moment of a molecule, the more vigorous is the oscillation in the microwave field.
The parameters defining the dielectric properties of the materials are dielectric constant (ε’)
and loss factor (ε’’). The dielectric constant gives the idea about the ability of a molecule to
be polarized by the applied field while the loss factor measures the efficiency with which the
microwave energy can be converted into heat. These two parameters depend on the frequency
of the microwave. The energy dissipation factor or loss tangent (tanδ = ε’’/ ε’) combines these
two parameters and gives idea of different materials to convert microwave energy into
thermal energy at given frequency. As Water is the predominant component of biological
materials, its presence directly enhances heating.
Extraction by microwave energy
In microwave extraction of biological active compounds from plant materials, the
microwaves reach the inner glandular, trichomes and vascular systems of the biological
PROCEDURE: MAE
1. Take measured amount (5 g) of crushed patchouli leaves in the flask.
2. Add specified amount of acetone (100 mL) in the flask and attach the
condenser for making open reflux system.
3. Set power at 300 W and time of operation at 10 min, and start the
operation.
4. After cooling the mixture, filter the solution.
5. Analyze the solution using gas chromatogrph. Dilute the solution if
necessary.
6. Calculate the amount of patchouli alcohol (PA) extracted using
calibration curve.
UAE
7. Take measured amount (2 g) of crushed patchouli leaves in the
beaker.
1. Add specified amount of acetone (50 mL) in the flask.
2. Insert the ultrasonic probe in the beaker.
3. Set the probe at 100 W power, 60 % cycle, 60 % amplitude and 10
min. of extraction time.
4. After extraction is complete, allow the mixture to cool and then filter
the solution.
