An initial step to do genetic recombination is of course to extract
DNA. In the succeeding exercise/lab activity you are expected to extract DNA from a selected organism.
MATERIALS:
1 banana Table salt
1 cup of water Ziplock bag Alcohol Empty plastic Filter paper glasses Dishwashing liquid Spoon PROCEDURE:
1. Cut the peeled banana
into small pieces and place it in the zip-lock bag.
2. Mash the banana by
pressing the bag with your hands. You could also use spoon in it.
3. Next we prepare our
solution. 4. Fill the glass with half a glass of water.
5. Add 1/2 tablespoon of
salt.
6. Add one tablespoon of
dishwashing liquid. Mix well.
7. Add the half solution
to the mashed banana. 8. Make sure to zip the bag properly. Mix well.
9. You can now filter the
mixture to the filter paper. Set aside.
10. Add the ice-cold
alcohol to the mixture.
11. Wait for 5 minutes
and observe the mixture.
We can now see the tiny DNA stand present in the mixture. CONCLUSION:
The experiment was quite an interesting learning
experience we had. We were bewildered at first as to how we should perform the task but we researched about banana extraction and took our time to understand it. When it was time for us to perform the experiment, there weren't much confusion about it but we had to do a second attempt because the first attempt wasn't a success. The second attempt was fortunately successful and we were able to extract the DNA of the banana. In conclusion, this experiment was very educational for us and it thought us a lot about DNA extraction. Teamwork made all the difference as well.
