Introduction:
Many things that are viewed using microscope particularly cell can
appear quite transparent under the microscope. The internal parts of the
cell, the organelles, are so transparent that they are often difficult to
see. Biologist have developed a number of stains help them to see the
cells and their organelles. In this experiment we will try to see what is
the structure of cell using an onion cell in a microscope.
Objective:
Identify the structure that is visible in an onion cell and it's shape
To know why onion is an easy experiment materis/tools
Materials:
Microscope
Microscope Camera
Microscope Slide
Knife
Toothpicks
Pipettes/ Dropper
Cover slips
Onion
Iodine Tincture
Cutting board
Procedures:
1. Preparing the slides
2. Wash and peel the onion
3. Slice the onion as thin as possible
(Thin layer of skin around the onion work best)
4. Clean your microscope slide with an alcohol wipe
(Do the same with cover slip)

5. Place your pieces of onion onto the slide and reposition them with
toothpick
6. A drop of Iodine onto the onion
7. Drop on the cover slip( apply pressure to coverslip but not enough to
break it.
8. Now your slide is complete
9. Observe under microscope
Analysis:
1. What is your observation ? Can you paste or draw the result of the
experiment.
-I observe that there are large, rectangular interlocking cells,
Clearly visible distinct cell walls surrounding the cells,Dark
stained nucleus, Large vacuoles at the center and Small granules
may be observed inside the cells (within the cytoplasm)
vacuole are indicators that help identify plant cells, such as seen
in the onion peel.
2. What are the shape of an onion cell?
- The shape of an onion cell is square or rectangular. The interior
of the cell is easy to see and focuses well for microscopic
observation.
3. In your opinion, why onion is used in an experiment ?
-An onion is made up of layers that are separated by a thin
membrane. For this experiment, the thin membrane will be used to
observe the onion cells. It can easily be obtained by peeling it from
any layer of the onion using tweezers.
4. Why they use iodine tincture for onion cell?
-The onion rigid cell walls allows cells to viewed in the microscope
and the iodine dye is a red allowed it to be seen.
