1. Have clear petri dishes, bacteria, and other equipment
ready 2. Turn on Bunsen Burner. 3. To prepare nutrient agar, suspend 28g in 1L of distilled water. While stirring, bring to boil. Boil 3-5 mins. Sterilise by autoclaving at 121°C for 15 minutes. Allow to cool to 70°C then pour into sterile plates and allow gel to set. Store set plates on their lids (upside down) in the fridge. || Add Mueller Hinton Agar powder to distilled/deionized water and bring to 1.0L. Mix thoroughly. Gently heat and bring to boiling. Distribute into tubes or flasks. Autoclave for 15 min at 15 psi pressure–121°C. Pour into sterile Petri dishes or leave in tubes. 4. Pour 30 ml of prepared agar into petri dishes. Do this until 75 petri dishes have been filled. Wait 45 minutes for agar to solidify. 5. Squeeze lemon juice out, and pour into a container. Dip 50 blank discs into it, and store them in container 6. Squeeze Onion juice out, and pour into container. Dip 50 blank discs into it, and store them in container 7. Using micropipette, pipette 100 ul of bacteria onto petri dish. Use bacteria spreader to spread it around 8. Place 4 disks on each petri dish. 9. Cover with parafilm 10. Incubate at 30 degrees for 24-72 hours
