Introduction
A cell is the smallest unit of life that can survive and reproduce on its
own, given information in DNA, energy inputs, raw materials, and
suitable environmental conditions.
‫ مع اعطاء‬،‫الخليه هي اصغر وحده في الحياه يمكنها البقاء والتكاثر من تلقاء نفسها‬
‫معلومات في الحمض النووي ومدخالت الطاقه واملواد الخام والظروف البيئيه املناسبه‬
• All cells are similar in three respects
‫جميع الخاليا متاشبهه في ثالثه جوانب‬

Materials Required ‫املواد املطلوبه‬
 ‫اعداد شريحه خليه البصل‬

The Preparation of onion cell slide

1. Pour some distilled water into a
‫صب بعض املاء املقطر في الزجاج الساعه‬
watch glass.

2. Peel off a leaf from half a piece of

onion and using the forceps, pull out
a piece of transparent onion peel
‫ اسحب قطعه من قشر البصل الشفاف ) البشره( من‬،‫تقشر ورقه من نصف قطعه من البصل باستخدام امللقط‬
(epidermis) from the leaf. ‫الورقه‬

3. Put the epidermis in the watch

glass containing distilled water.
‫ضع البشره في زجاج الساعه املحتوي على املاء املقطر‬

4. Take a few drops of safranin
solution in a dropper and
transfer this into another watch
glass. ‫خذ بضع قطرات من املحلول الزعفران في قطاره رانقلها الى زجاج ساعه اخر‬

5. Using a brush, transfer the

‫ انقل القشر الى زجاج الساعه الذي يحتوي على محلول الزعفران‬، ‫باستخدام فرشاه‬
peel into the watch glass
containing the safranin solution.

6. Let this remain in the Safranin

solution for 30 seconds, so that
the peel is stained.

‫حتى يتلطخ القشر‬، ‫ ثانيه‬٣٠ ‫اتركه في محلول الزعفران ملده‬

7. Take the peel from the
Safranin solution using the
brush and place it in the watch
glass containing the distilled
water.
8.Take a few drops of glycerine
.‫ قطرات في وسط شريحة زجاجية جافة‬3-2 ‫ خذ بضع قطرات من الجلسرين في قطارة واسكب‬-8
in a dropper and pour 2-3 drops
at the center of a dry glass slide.

9.Using the brush, place the

peel onto the slide containing
glycerine.

10. Take a cover slip and place it

gently on the peel with the aid of

a needle.

11. Remove the extra glycerine

using a piece of blotting paper.

12. Place this glass side

 Observations
.‫• يوجد عدد كبير من الخاليا املنتظمة الشكل ترقد جنبًا إلى جنب ولكل خلية جدار خلوي مميز‬
• There are a large number of
regularly shaped cells lying side by
side and each cell has a distinct cell
wall.
.‫• توجد نواة مميزة على محيط كل خلية‬
• A distinct nucleus is present on the
periphery of each cell.
.‫• لوحظ السيتوبالزم امللطخ بشكل خفيف في كل خلية‬
• Lightly stained cytoplasm is
observed in each cell.

• A large vacuole is present at the

centre of each cell, and is
surrounded by the cytoplasm.
.‫ ويحيط بها السيتوبالزم‬، ‫• توجد فجوة كبيرة في وسط كل مرأة‬
 Conclusion
• As cell walls and large vacuoles
are clearly observed in all the
cells, the cells placed for
observation are plant cells.
‫• نظرًا ألنه يتم مالحظة جدران الخاليا والفجوات الكبيرة‬
‫ فإن الخاليا املوضوعة للمراقبة‬، ‫بوضوح في جميع الخاليا‬
.‫هي خاليا نباتية‬

Precautions
.‫• استخدم فرشاة لنقل القشر من جهاز إلى آخر‬
• Use a brush to transfer the peel from one apparatus to another.
.‫• يجب أال يكون تلطيخ القشر داكنًا جدًا أو فاتحًا جدًا‬
• Staining of peel should neither be too dark, nor too light.
• Extra glycerine stain should be removed using blotting paper.
.‫• يجب إزالة بقعة الجلسرين الزائدة باستخدام ورق نشاف‬
