( Alginic acid Extraction )

Determination of Alginic acid content in Seaweed Powder AS Sodium Alginate

(By Vishweswara Rao and Mody ,1965.)

Reagents:

2% Sodium Carbonate

10% Formalin

10 % Sodium Hydroxide

Commercial Seaweed Powder /flakes

Ethanol

Distilled water

Instruments / Glass wares required

Conical flasks

Magnetic stirrer

Centrifuge

Vacuum filtration unit

Hot air oven

Petri plates

Weighing scale

Procedure:
Sodium alginate was extracted from the selected seaweeds by the modified method based on the study of

Torres et al.

1. About 10 g of powdered samples were immersed in 10% formaldehyde for 24 h, in an airtight

conical flask.

2. Then, the solution was filtered out using a Glass Funnel and Whitman qualitative filter paper (70

mm), while the soaked seaweeds were washed with distilled water two to three times.

3. The HCl solution (0.2 M) was added and maintained at room temperature for 24 h.

4. The solution was separated by filtration, and the residue was washed with distilled water two to

three times.

5. The alginate was extracted with the addition of sodium carbonate (2% m: v) to the residue

previously obtained, and this process occurred overnight.

6. Then, the extract was filtered through a gauze cloth bag (Musclemen cloth) and the filtrate was

bleached with 2.5% (v: v) sodium hypochlorite.

7. The extracted sodium alginate was precipitated from the solution by ethanol, then washed with

acetone, and dried overnight at 60 ◦C.

8. The alginate yield was measured as follows:

Yield of sodium alginate (%) = Weight of alginate /Weight of seaweed dried biomass X 100
