[2]-Define DNA sequence, Chromosome libraries and Genomic

library:
DNA sequence A clone from a genomic library, or any other clone, can be analyzed
to determine the nucleotide sequence of the DNA insert, as well as to determine the
distribution and location of restriction sites. Its nucleotide sequence is the most
detailed information one can obtain about a DNA fragment. The information is useful,
for example, in computer database analyses for comparing sequences from different
genomes, which can tell us how closely related two organisms are, or for identifying
gene sequences and the regulatory sequences—like promoters, silencers, and
enhancers—that control gene expression.(Hartog et al., 1967)
A library consisting of a collection of cloned DNA fragments derived from one
chromosome is called a chromosome library.(Hartog et al., 1967)
A genomic library is a collection of clones that contains at least one copy of every
DNA sequence in an organism’s genome.(Hartog et al., 1967)

[4]-List factors increase and decrease intracellular free cholesterol

concentration:
Cholesterol balance in tissues: Many factors will determine the cholesterol balance at
the cellular level.
 Increasing:

 increase is due to uptake of cholesterol-containing lipoproteins by receptors

 the LDL receptor or the scavenger receptor.
 uptake of free cholesterol from cholesterol-rich lipoproteins to the cell
 hydrolysis of cholesteryl esters by the enzyme cholesteryl ester hydrolase
 Phosphatases dephosphorylate (fed state) and activate HMG-CoA activity;
therefore increasing cholesterol.
 Increased synthesis of cholesterol.(Murray & Davis, 2003)

 decreasing:

 Decrease is due to efflux of cholesterol from the membrane to HDL

2
 promoted by LCAT (lecithin: cholesterol acyltransferase)
 esterification of cholesterol by ACAT (acy l- CoA: cholesterol
acyltransferase)
 utilization of cholesterol for synthesis of other steroids, such as hormones, or
bile acids in the liver.
 inhibation synthesis of cholesterol.
 down-regulates synthesis of the LDL receptor.(Murray & Davis, 2003)

[5]-Describe metabolic fate of cholesterol inside the body:

[6]-Compere between Carcinoma and sarcoma:

Carcinoma Sarcoma
Define Cancers that grow in soft the Cancers that grow in the epithelial
epithelial cells Less common
connective or supportive tissues, more
common
Example Adenocarcinoma ,squamous Osteosarcoma (bone) and
cell carcinoma ,anaplastic chondrosarcoma (cartilage)
carcinoma

Aga More above 45 More in yong adult

Rate of growth Slower than sarcoma Very rapid
Frequency common Rare
2
Spread Early by blood sarcoma Early by lymphatic's
Prognosis Less worse More worse
Blood supply less Rich with hemorrhage and necrosis
And blood vessels
Histology Cells arranged in groups Cells arranged individual
Premalignant phase May have a pre-malignant Not believed to have a pre-malignant
phase phase
Striking Age group Generally, below fifty Generally , above fifty
Form and consist Usually hard Soft or fleshy
Commend side Any where lung liver bone Lung
brain
Radio sensitivity More sensitivity Less sensitivity
