Gram Stain Report
Gram Stain:
Gran stain is a technique used in labs to distinguish gram-positive eubacteria
from gram-negative eubacteria.
Materials Used:
Crystal violet
Iodine
Alcohol
Safranin
Water
Tweezer
Medical gloves
Heat source
Eye dropper
Gram-positive bacteria (in agar solution)
Gram-negative bacteria (in blood agar solution)
Microscope
Slides
The Procedure:
We started the procedure by heating the tweezer in order to become
sterilized.
Then we put a drop of water onto a slide.
We put both gram-positive and gram-negative bacteria onto a slide by
using a tweezer then heated the tweezer to kill any remaining bacteria on
the tweezer to prevent harm, and damage to other people.
We dropped Crystal violet on the slide by using eye dropper, then washed
the slide with water after 30 seconds.
We put iodine on the slide then washed the slide after 1 minute.
We put Alcohol on the slide and washed it after 10 seconds.
We put Safranin on the slide which had a brown-reddish color, then
washed it after 1 minute.
Then we put the slide under a microscope to observe it closely.
Result:
After completing the procedure and looking at the slide under a light-
microscope, we observed that most of the bacteria we had were gram-negative
and their color were pink but we also saw some purple colored spherical
bacteria under the microscope which was the gram-positive bacteria.
